血管性痴呆和Alzheimer病的载脂蛋白E基因多态性研究

摘　　要：目的　探讨载脂蛋白 E(APOE)基因多态性与老年期痴呆 [血管性痴呆 (VD)、Alzheimer型痴呆(AD) ]的相关性。方法　采用聚合酶链反应限制长度片段多态性技术 (PCR- RFL P)检测 2 6例 VD(下称 VD组 ) ,32例 AD(下称 AD组 ) ,4 0例正常人 (下称对照组 ) APOE基因型和等位基因频率并进行对比分析。结果　三组均以 ε3/3为优势基因型。 VD组、AD组和对照组分别为 5 0 .0 0 %、5 6 .2 5 %、80 .0 0 % ,VD、AD两组较对照组有显著性差异 (P<0 .0 5 )。ε3/4基因型在 VD组、AD组分别为 7%、9% ,较对照组 (3% )也有显著性差异 (P<0 .0 5 )。ε4 /4基因型在各组中均在 5 %以下。 VD和 AD患者等位基因ε3降低、等位基因ε4增高 ,与对照组相比较差异有显著性 (P<0 .0 5 )。结论　基因型 ε3/4与 VD、AD具有相关性 ,等位基因Objective To explore the relationship between polimorphisms in apolipoprotein E gene and vascular dementia or Alzheimer’s diseases.Methods We collected 26 patients with vascular dementia(VD),32 patients with Alzheimer’s diseases and 40 cases of normal Chinese Han nationality.Exon 4 of apolipoprotein E fragment including coding for 112 and 158 aminoacid residues was amplified by PCR.The genotype and its alleles frenquencies were calculated and compared.Results The ε 3/3 genotype of apolipoprotein E in patients with VD and AD,also in control group was dominant character.The frequencies of ε 3/3 genotype in VD and AD was lower than that in control(P<0.05),50.00% in patients with VD,56.25% in patients with AD and 80.00% in control group.While ε 3/4 genotype in patients with VD and AD were higher than that in control group(P<0.05).The ε 4/4 genotype in 3 groups were all lower (not more than 5%).APOE alleles ε 3 in patients with VD and AD was lower than that in control and ε 4 was higher (P<0.05).Conclusion Apolipoprotein ε 3/4 genotype may contribute to the development of patients with vascular dementia or Alzheimer’s diseases.ε 4 allele is probably a genetic susceptible factor of both vascular dementia and Alzheimer’s diseases.
