摘 要:目的 研究不同诱导分化剂单独或与热休克联合作用下 ,K562细胞JWA蛋白表达的特点和规律 ,探讨JWA与热休克蛋白 70 (Hsp70 )表达的关系以及可能涉及的信号调节机制。方法 建立诱导K562细胞定向分化以及和热休克联合作用的实验模型 ,用Westernblot方法检测JWA、Hsp70、热休克转录因子 1(HSF1)、HSF2等蛋白表达。结果 (1)佛波酯 (TPA ,10 0、2 0 0ng/ml)或阿霉素 (adri amycin ,4× 10 - 8mol/L)处理细胞 48h后 ,再热休克 (42℃ )处理 2h ,JWA和Hsp70蛋白表达均增加 ;而氯化高铁血红素 (hemin ,3× 10 - 5 mol/L ,48h)、阿糖胞苷 (Ara C ,80ng/ml,48h)和三氧化二砷 (As2 O3,1× 10 - 6 mol/L ,48h)处理后再热休克处理细胞 2h ,则JWA和Hsp70蛋白表达均下调 ;全反式维A酸(ATRA ,1× 10 - 6 mol/L ,48h)处理细胞后 ,再热休克处理 ,则JWA和Hsp70蛋白表达无明显变化。 (2 )不同诱导分化剂处理K562细胞后再热休克处理 ,HSF1和HSF2表达变化均不明显。结论 JWA不但参与K562细胞的诱导分化调节 ,也参与细胞对热应激的调节 ,JWA可能是多种化学诱导分化剂和热休克的共同靶基因。诱导K562细胞分化过程中 ,Hsp70的表达不需要热休克转录因子介导 ,JWA可能是一种与Hsp70信号通路平行的新的信号分子。ObjectiveTo study how the combined effects of various differentiation inducers and heat stress on the expression of JWA pr otein in K562 cell,the relationship between JWA and Hsp70 expression,and the sig nal regulation mechanism possibly involved. MethodsThe e xperimental model was established in K562 cells.Various directional differentiat ion inducers(TPA,Ara-C,hemin,adriamycin,ATRA and As 2O 3) were used alone or combined with heat shock treatment(42 ℃,2 h).Western blot was used for detecti ng expressions of JWA,Hsp70,heat stress factor 1(HSF1) and HSF2. Resu lts(1)The expressions of both JWA protein and Hsp70 were significan tly up-regulated after K562 cells treated by TPA(100,200 ng/ml) or adriamycin(4 ×10 -8 mol/L)48 h,and followed by heat shock(42 ℃,2 h).However,the opposi te effects were observed when the cells treated by hemin (3×10 -5 mol/L, 48 h),Ara-C(80 ng/ml,48 h) and As 2O 3(1×10 -6 mol/L,48 h) followed by 2 h heat shock.No obvious changes were found when the cells treated by ATRA(1×10 -6 mol/L,48 h) alone or followed by heat shock.(2)Both the heat shock trans criptional factors HSF1 and HSF2 did not show any significant changes when K562 cells were treated with various differentiation inducers and followed by heat st ress. ConclusionJWA not only takes part in the regulatio n of K562 cellular differentiation,but also of heat stress,it might be the co-t arget gene of several differentiation inducers and heat stress.The expression of Hsp70 seems not mediated by both HSF1 and HSF2 in K562 cells undergoing directi onal differentiation or heat stress treatment.JWA is likely to be a new signal m olecule similar to Hsp70 signal pathways.The results show that JWA takes part in the mechanism of K562 cell response to heat stress.
