摘  要:目的 观察新辅助化疗对宫颈鳞癌细胞增殖及凋亡的影响。方法 对 2 4例宫颈鳞癌采用以顺铂为主联合羟基喜树碱、博来霉素方案进行术前的新辅助化疗 ,对化疗前后的宫颈鳞癌组织分别进行如下检测 :以流式细胞术检测组织中PCNA、Ki- 6 7的表达 ;运用TUNEL法检测肿瘤细胞的凋亡指数 (AI)。结果 化疗前宫颈鳞癌组织中PCNA、Ki- 6 7的表达及AI检测值分别为 0 .386± 0 .0 78、0 .5 5 0± 0 .12 7、0 .0 5 2± 0 .0 2 7,1个疗程结束后 3d重新取材检测的结果分别为 0 .12 5± 0 .0 4 0、0 .2 37± 0 .0 76、0 .2 4 8± 0 .0 78,化疗前后三者的检测值有显著差异 P <0 .0 1,统计值分别为t =2 2 .85 9、t =15 .813、t =16 .0 6 ;化疗后PCNA、Ki- 6 7的表达量下降分别为 0 .2 6 2± 0 .0 5 6、0 .313± 0 .0 97,化疗后AI表达高于化疗前 0 .196± 0 .0 6 0 ,三者表达变化程度之间无相关性P>0 .0 5。结论 对宫颈鳞癌进行顺铂联合羟基喜树碱、博来霉素的新辅助化疗能显著抑制肿瘤细胞增殖、增加凋亡 ,PCNA、Ki- 6Objective To determine the effect of neoadjuvant chemotherapy(NAC) for cervix cancer on cellular proliferation and apoptosis of cervical cancer.Methods 24 patients with squamous carcinoma of the uterine cervix, were treated with one cycles of cisplatin combined with hydroxycamptothecine 、bleomycin regiman . The expression of PCNA、Ki-67 and apoptosis index(AI) were detected before and after NAC ,with flow cytometry and TUNEL. Results Before NAC the expression of PCNA、Ki-67 and AI were 0.386± 0.078、 0.550± 0.127、 0.052± 0.027 respectively, and 3 days after NAC which were 0.125± 0.040、 0.237± 0.076、 0.248± 0.078. Significant difference could be observed before and after NAC treatment (P<0.01),(t=22.859、t=15.813、 t=16.06),The expression of PCNA、Ki-67 decreased with 0.262±0.056、0.313±0.097 after NAC, while AI increased with 0.196±0.060,but no significant correlation was existed among the changing of three group(P>0.05) . Conclusion NAC with HDB combination could inhibit proliferation and hasten apoptosis of tumor cell in cervical squamous carcinoma tissue. And the changing of expression of PCNA、Ki-67 and AI maybe one of molecule mechanism leading to anti-tumor function of NAC.
