摘 要:作者采用体外集落培养法研究了新卟啉类光敏剂癌光啉加光照对人白血病细胞系的体外选择性杀伤作用。加癌光啉10μg/ml,体外37℃孵育1.5h,照光4min对U937、HIMeg和HL60等白血病细胞系和HL60细胞与正常人MNC混合细胞中的HL60细胞集落的杀伤均大于4个对数级(log),而对正常CFU-GM的杀伤仅为0.63±0.59 log。说明癌光啉加光照确能选择性杀死白血病细胞,可用于自身骨髓移植治疗白血病时净化骨髓。探讨了影响光敏杀伤效应的几个因素,为临床净化处理大量骨髓细胞提供了实验依据。One of the important problems of using autologous bone marrow transplantation in the treatment of leukemia is to purge residual leukemic cells from the autologous marrow grafts in vitro. This paper reported the selective killing effect of photocarcinorin (PsD-007), a new photosensitive agent from porphyrin, plus light on human leukemic cell lines. It was observed that photocarcinorin (10mg/L) and 4 minutes of daylight fluorescence irradiation could eliminate 1.61 log, 4.52 log, >4.25 log, > 5.16 log of K562, U937, HIMeg and HL60 cells, respectively, but spare 23.37 ± 25.66% of normal CFU-GM (0.63 log killing). When HL60 cells mixed with normal mononucleated bone marrow cells in 1:9 ratio were studied, the killing of HL60 colonies was more than 4.29 log. The result suggested that photocarcinorin plus light could eliminate leukemic progenitors, and might be used in clinical ABMT. Several factors that might affect the photosensitization effect on cells were discussed.
