人颊癌细胞在胶原凝胶中生长和浸润特性初步研究

摘　　要：探讨胶原蛋白凝胶作为颊癌细胞在体外培养环境下生长和浸润模型的价值。方法　从鼠尾肌腱中提取出较纯的Ⅰ型胶原蛋白 ,制成胶原蛋白凝胶体 ,将建株人颊癌细胞置于其中培养生长 ,从形态学特征上初步探讨胶原蛋白凝胶模型的价值。结果　从鼠尾肌腱提取的Ⅰ型胶原蛋白 ,能形成胶原蛋白纤维束网状结构—胶原蛋白凝胶 ;其内生长的颊癌细胞所处的生长环境 ,类似于机体内的生长环境 ,颊癌细胞有很高的生长率。结论鼠尾肌腱胶原蛋白凝胶 ,能制成理想的肿瘤细胞的生长及浸润模型 ;建株颊癌细胞能在立体胶原凝胶中生长 ,保持着细胞原有的形态特征及生物学特性。Objective To search the effects of the invasive growth model of collagen gel for studying culture model of human buccal squamous carcinoma cell line in vitro.Methods Collagen Ⅰ was extracted from rat tail tendons by using a simple and economical method,from which collagen gel netted construction was made,and then human buccal squamous carcinoma cell line was cultured in the gel.Results Collagen Ⅰ extracted from rat rail tendon could form collagen gel netted construction.Growing condition of human buccal squamous carcinoma cell line was similar to that of human being.Human baccal carcinoma cell had a high growing rate in the collagen gel.Conclusion Collagen gel from rat tail tendon can form perfect growing and invasive model of tumor cells;human buccal squamous carcinoma cell line can grow in three-dimensional collagen gel and keep the original morphological features and biological properties.
