摘 要:目的 :探讨在高糖环境下人肾小球系膜细胞与内皮细胞间相互作用对TGF β1 mRNA表达的影响和茶多酚的干预作用。方法 :建立系膜细胞和内皮细胞共培养模型 ,分空白对照组、高糖组、茶多酚干预组和茶多酚对照组 ,培养 0、12、3 6h后 ,应用RT PCR法检测模型中两种细胞转化生长因子 β1 (TGF β1 )的表达情况。结果 :高糖可上调共培养模型中系膜细胞和内皮细胞TGF β1 mRNA表达 ,其作用显著强于单独培养的系膜细胞和内皮细胞 ,茶多酚可降低共培养模型中细胞TGF β1 mRNA表达 ,且效果优于单独培养。结论 :(1)高糖环境下系膜细胞和内皮细胞间存在相互作用 ,这种作用能促进两种细胞TGF β1 mRNA高表达。 (2 )茶多酚通过对TGF β1 mRNA表达的抑制影响细胞间相互作用。Objective To study the effect of high glucose and tea polyphenols (TPs) on the interaction of human mesangial cells and endothelial cells. Methods Mesangial cells and endothelial cells were cultured alone or together in high glucose media with or without TPs for 0-h, 12-h and 36-h respectively. The ex pressi ons of TGF-β 1-mRNA in the cells were measured by RT-PCR. Resul ts High glucose i ncreased the expression of TGF-β 1-mRNA in co-cultured mesangial cells as well as in co-cultured endothelial cells and the effect became more prominent than in sing le-cultured cells,which could be interruptted by TPs more effectively in co-cu ltured cells than in single-cultured cells. Conclusions (1)There is some kind of interaction betwee n human mesangia l cells and endothelial cells in high glucose condition, and the interaction can increase the expression of TGF-β 1-mRNA. (2)TPs inhibit the expression of TGF-β 1-mRNA by affecting the intercellular interaction between the mesangial c ells and the endothelial cells.
