摘  要:目的 观察静脉注射骨髓间质干细胞 (MSC)对血管性痴呆 (VaD)模型大鼠认知功能的影响。 方法 体外分离培养、扩增成年大鼠MSC。利用结扎双侧颈总动脉方法制备慢性前脑缺血动物模型 ,造模后 6h采用静脉注射Hoechst33342荧光标记的MSC(6× 1 0 6/ 0 5ml) ,观察注射后 30、6 0d大鼠脑内荧光标记细胞数及分化的功能细胞和认知功能变化。 结果 与正常对照组相比 ,VaD大鼠主动回避反应正确率 (90 2± 4 8、37 6± 9 3、4 3 5± 1 0 5 )min、游泳时间 (0 39± 0 1 8、2 47± 1 2 2、1 89± 0 90 )min、错误次数 (1 5 5± 0 40 ,4 5 6± 1 6 3、3 48± 0 6 6 )和盲端时间 (0 1 4± 0 0 7、1 40± 1 1 4、0 87± 0 76 )min ,在造模后 30d和 6 0d差异有显著性 (P <0 0 1 )。静脉注射MSC后 30d和 6 0d ,VaD大鼠认知功能显著改善 ,但 6 0d组Hoechst33342荧光阳性细胞 (2 6 34 2 6± 2 81 73)以及免疫荧光双标细胞数较 30d组 (375 9 45± 2 6 8 82 )明显减少 (P <0 0 1 )。 结论 静脉注射MSC可显著改善VaD大鼠认知功能 。Objective To explore the effects of intravenous infusion of marrow stromal cells(MSCs) on cognitive function of vascular dementia rats Methods Marrow stromal cells(MSCs) were isolated and cultured from adult rats Test rats were subjected to the permanent bilateral carotid arteries ligation (BCAL) for 2 months.Test group received intravenous infusion of 6×10 6 /0 5 ml of MSCs which were fluor prelabelled with Hoechst 33342 in vitro at 6 hours after BCAL. Cognitive function(active avoidance response and water maze test) were performed before and 30,60 days after BCAL. Immunohistochemistry was used to identify MSCs or cells derived from MSCs in brain. Results Active avoidance response, swimming time, number of errors and error time of VaD rats were significantly different from normal control group( P <0 01). Significant improvement of cognitive impairment were found in VaD rats infused with MSCs at 30 and 60 treatment days compared with VaD groups. MSCs survived and localized to the brain tissue.The numbers of Hoechst33342 immunohistofluorescence positive cells and double immunostaining cells were significantly decreased in 60 days group compared with 30 days group( P <0 01). Conclusions MSCs delivered to ischemic brain tissue through intravenous route improve the cognitive impairment of VaD rats, and MSCs may survive and differentiate in brain tissue of VaD rats.
