摘　　要：目的 :研究 TRF1蛋白在肾脏恶性肿瘤及相应癌旁组织中的表达水平。方法 :定量分析肾组织中 TRF1蛋白的表达水平 ,应用经倍比稀释的 TRF13 3 -2 77纯化蛋白作为定量标准 ,建立了以抗 TRF13 3 -2 77单抗作定量 Wes-tern- blot的方法 ,并以该方法检测 TRF1蛋白在组织中的表达水平。结果 :32例肾肿瘤样本均不同程度的表达TRF1蛋白质 ,均值为 2 .4 2 8± 1.35 2 μg/ μl,癌旁自身对照组织 TRF1的表达水平为 3.6 11± 1.92 2 μg/ μl(t=5 .776 ,P<0 .0 0 1)。肿瘤组织内 TRF13 3 -2 77表达水平较相应癌旁组织显著降低 ,并与肿瘤的恶性程度相关 ,差异具有显著性意义 (P<0 .0 1)。结论 :TRF1蛋白在肾脏恶性肿瘤中的表达明显减低 ,并与肿瘤恶性程度呈负相关。Objective: To investigate the expression levels of telomere repeat binding factor1(TRF1) protein in normal kidney tissue and kidney cancer. Methods: Specimens of kidney cancer and pericancerous tissues were collected from 32 cases of renal carcinoma.A quantitative Western blotting technique was developed using TRF1 mo noclonal antibody to determine the expression level of TRF1 protein in total protein extracts from tissue specimens. Results: The expression level of TRF1 protein was higher in normal kidney tissues (3 611±1 922 μg/μl) than that of cancer tissues (2 428±1 352 μg/μl) ( t=5 776,P <0 01). Conclusion: The expression level of TRF1 protein is significantly reduced in kidney cancer and the level is negatively correlated with malignant degree of the cancer. [
