摘  要:目的 探讨白细胞介素 1 β(IL 1 β)致痫或促痫的细胞内信号转导机制。 方法 动物行为学观察、免疫细胞化学方法和放射免疫分析法。 结果 行为学观察表明 :IL 1 β组大鼠有轻至中度癫痫发作 ,IL 1 β和谷氨酸(Glu)联合注射组有中至重度癫痫发作 ,IL 1受体拮抗剂 (IL 1ra)预处理可减轻或抑制癫痫发作。IL 1 β致痫或促痫时 ,大脑皮质和海马神经元Gas蛋白免疫反应明显增强 ,海马组织cAMP含量明显增高 ,IL 1ra预处理 ,则可抑制cAMP的升高。 结论 IL 1 β可能有致痫和促进Glu致痫作用 ,IL 1ra可抑制此作用。IL 1 β致痫或促痫的细胞内信号转导机制在海马区与Gas蛋白介导的AC cAMP途径可能有密切关系。Objective To investigate the mechanism of intracellular signal transduction of seizure induced by interleukin1β(IL1β). Methods Animal behaviour observation,immunocytochemical staining and radioimmunoassay(RIA) were performed. Results The behavior of rats displayed light to middle seizure in IL1β group,middle to heavy seizure in united group of IL1β and Glu,but only light or no seizure in pretreatment group by IL1 receptor antagonist(IL1ra).Gas immunoreaction was obviously increased in cerebral cortex,hippocampus comparing with the control group.The level of cAMP in hippocampus was raised in the IL1β group and the united group of IL1β and Glu.Pretreatment of IL1ra could inhibt the raise of level of cAMP.Conclusion IL1β could induce or promote seizure.The pretreatment of IL1ra could inhibit or lighten seizure.The intracellular signal transduction mechanism of IL1βinduced epilepsy might be closely related to G protein and ACcAMP pathway in hippocampus.;
