摘 要:目的 研究人胚肺成纤维 (HEL)细胞感染人巨细胞病毒 (HCMV)后 ,转录激活蛋白 1(AP 1)活化和原癌基因c junmRNA表达的时序性变化 ,探讨其在HCMV感染中的作用。方法 采用原位杂交法检测c junmRNA的表达 ,凝胶电泳迁移率改变试验 (EMSA)检测AP 1活性。结果 HEL细胞感染HCMV后 15min ,c junmRNA即有阳性表达 ,30~ 6 0min达高峰 (P <0 0 1) ,以后逐渐下降 ,正常对照组HEL细胞无c junmRNA表达。HCMV感染后AP 1亦迅速被活化 ,至 2h达高峰 (P <0 0 1) ,其后有所下降 ,维持于一定水平。结论 HEL细胞感染HCMV后 ,c junmRNA表达迅速增加 ,AP 1活化。提示HCMV可能通过刺激原癌基因过度表达而参与细胞的信号传导 ,以干扰细胞增殖和分化的调控 ;AP 1活化为早期病毒基因有效表达所必需 ,并可启动一系列前炎性细胞因子的基因转录与蛋白合成。Objective To investigate the scheduling change of the activation of AP-1 and the expression of proto-oncogene c-jun mRNA in HCMV-infected HEL cells so as to explore the role of c-jun and AP-1 in HCMV infection. Methods The expression of c-jun mRNA was detected by in situ hybridization (ISH) and activity of AP-1 was measured by electrophoretic mobility shift assay (EMSA).Results The expression of c-jun mRNA was detected in the infected HEL cells at 15 min post infection, reached the peak at 30—60 min (P<0.01), then slowed down, while there was no expression of c-jun mRNA in the uninfected HEL cells. AP-1 was also activated rapidly post HCMV infection and reached the highest level at 2 h (P<0.01), then declined to some extent and maintained at a certain level. Conclusion Post HCMV infection, the expression of c-jun mRNA was increased and AP-1 was activated. It suggested that HCMV might participate in the signal transduction via stimulating the overexpression of proto-oncogene and the activation of AP-1 may be necessary for the early efficient expression of HCMV gene.
