4-1BBL逆向信号促进单核细胞的增殖和分泌细胞因子的作用

摘  要:采用抗人 4 1BBL单克隆抗体 1F1包被 2 4孔培养板 ,加入经免疫磁珠阴性选择获得的高纯度人外周血单核细胞 ,动态观察细胞的生长状态 ,并用3 H TdR掺入法分析单核细胞增殖 ,应用ELISA动态测定培养上清中IL 6、IL 10、M CSF和FL等细胞因子水平。结果发现 1F1非常有效地促进单核细胞的增殖 ,1F1组单核细胞分泌高水平的M CSF以及IL 6和FL增加 ,但 1F1组单核细胞分泌IL 10水平低于对照组 (P <0 0 5 )。由此表明 ,抗人 4 1BBL单克隆抗体 1F1通过激发单核细胞 4 BBL逆向信号 ,介导单核细胞分泌M CSF、IL 6和FL。上述细胞因子联合作用 ,从而促进了单核细胞的增殖。To evaluate the effect of proliferation and cytokine secretion of freshly isolated monocytes stimulated by reverse signaling of 4-1BBL, monocytes were purified from PBMC by negative MACS selection and cultured in the 24 wells, which were pre-coated with anti-human 4-1BBL monoclonal antibody 1F1(1 μg/ml). The proliferative responses of monocytes were determined by 3H-TdR incorporation assay and cell number counting and the cytokine concentration in the supernatants of monocytes was assayed by ELISA. Experimental results showed that mAb 1F1 could effectively enhance the proliferation of monocytes from PBMC, high level M-CSF secreted by the monocytes in 1F1 mAb groups was detected, and the IL-6 and FL levels of 1F1 mAb groups increased obviously than the control groups(P<0.05). While IL-10 levels of 1F1 mAb groups were lower than murine IgG1 control groups. These data suggested that 1F1 mAb could mediate the secretion of IL-6, IL-10, M-CSF and FL by monocytes via 4-1BBL reverse signaling. It is speculated that combination of such cytokines might be involved in the mechanism of proliferation and immuno-modulation of monocytes.
