摘  要:以淀粉为原料,在特定条件下制备成一种可溶性水解液。经高效液相色谱等手段检测发现,内含纤维二糖等诱导物。实验证明,这种淀粉水解液能有效地诱导里氏木霉(Trichoderma reesei Rut C30)合成纤维素酶。在自由菌丝产酶条件下,滤纸酶活和纤维二糖酶活分别为1.51IU/mL和0.10IU/mL;采用固定化菌丝细胞在一批加料条件下产酶,滤纸酶活为1.30IU/mL,略低于自由菌丝产酶的结果,但纤维二糖酶活(0.19IU/mL)要比自由菌丝发酵高得多。作者认为,若采用分批添料技术,有利于消除因葡萄糖浓度过高对产酶形成的抑制作用,酶活可望进一步得到提高。The hydrolyzate derived from starch at special reaction conditions was found containing inducers such as cellobiose by high performance liquid chromatography. It was proved that so derived starch hydrolysate was able to induce cellulase production effectively by Trichoderma reesei RUT C30. When using free mycelia fermentation,the filter paper and cellobiase activities were 1. 51 IU/mL and 0. 10 IU/mL,respectively. In repeated batch fermentations using immobilized cells,the filter paper activity was 1. 30 lU/mL, and the cellobiase activity reached 0. 19 lU/mL,which was 1. 9 times as high as that of the free mycelia fermentation. To promote further cellulase activity, the technique of the fed-batch fermentation was recommended.
