摘  要:目的 采用免疫组化的方法探讨自体移植脾组织中神经纤维的再生过程。方法 健康Wistar大鼠 63只 ,体质量 10 0~ 12 0g ,分为实验组及假手术组 ,术后 7、14、3 0、60、90、12 0、180d取两组脾组织 ,行免疫组化ABC法抗NPY抗体染色、图像分析定量测定。结果 术后 3 0d内 ,自体移植脾组织无NPY+ 神经纤维 ;术后 60d ,NPY+ 神经纤维出现于移植脾组织周边区域并随时间推移向移植脾组织中央实质区伸展 ;术后 180d移植脾组织内NPY+ 神经纤维分布及免疫组化阳性染色区域密度接近正常。结论 应用免疫组化方法对自体脾组织移植后神经纤维再生过程进行研究 ,可清晰显示神经纤维再生规律 。Objective To study the regeneration of nerve fiber in autotransplanted spleen tissue by using immunohistochemical technology. Methods A total of 63 healthy Wistar rats were randomly divided into splenectomy and splenic autotransplantation group and sham operation group. Regeneration process and density changes of the regenerated nerve fibers in splenic autotgrafts of the two groups were studied by immunohistochemical methods and computer image analysis on day 7, 14, 30, 60, 90, 120 and 180 postoperatively. Results The regenerated nerve fibers had no regeneration on day 30 postoperatively, but on day 60 nerve fibers appeared in the peri region of the autotransplanted spleen and extended into the inner part of splenic autotransplants gradually. Density of the nerve fibers gradually became greater and almost normal 180 days after operation. Conclusion The regenerated nerve fibers could be revealed clearly by using immunohistochemical technology in autotransplanted spleen. This technology will be the important support for the research work of splenic autotransplantation.
