摘  要:目的 探讨A5 4 9肺腺癌多细胞球体内照射合适的3 2 P吸收剂量 ,以便临床对肺癌实体瘤作间质治疗选择核素内照射吸收剂量时给予参考。方法  96孔盘每孔一个球体 ,15Gy3 2 P内照射 2 4h后当天用直线加速器产生的 6MV高能X线作另外一组球体的连续 15Gy外照射 ,剂量率 2Gy/min。照射结束后第 8和第 15d用3 H 脱氧葡萄糖 (3 H DG)和3 H胸腺嘧啶核苷 (3 H TdR)摄取实验 ,流式细胞仪、光镜和电镜观察。结果 ① 15Gy内、外照射结束 8d后3 H DG摄取明显低于对照组 (P <0 .0 5 ) ,外照射组低于内照射组 (P <0 .0 5 ) ;3 H Tdr摄取低于对照组 (P <0 .0 1) ,内、外照射组比较P =0 .736 ;15d后 ,内、外照射组和对照组3 H DG摄取比较P >0 .0 5 ,照射组的3 H Tdr摄取低于对照组 (P <0 .0 5 ) ,照射组之间无显著差异 (P =0 .995 )。②内、外照射结束 1周后 ,外照射组的G2 M %和凋亡率高于内照射组 ,2周后比较两者差别不大。③内照射后球体未明显生长 ,外照射组略大于内照射组 ;电镜观察到坏死和凋亡 ,外照射多于内照射。结论 同等剂量照射时外照射的杀伤力更大 ,为3 2Purpose: To compare the treatment effect between internal and external irradiation in A549 Multicellular Spheroids(MTS) so that a conference could be acquired when the interstitial treatment is performed in lung cancer. Methods: Every MTS was placed in every hole of 96 holes plate, in the same time after 24 hours of internal irradiation with 15 Gy of Na2H32PO 4 was done, another group of MTS were received 15 Gy of 6 MV external irradiation produced by liner accelerator, with the radiation rate 2 Gy/min. In the eighth and fifteenth days after irradiation, uptake test with 3H-deoxyglucose(3H-DG)and 3H-TdR, flow cytometry, microscope and electron microscope were used to monitor the treatment effect. Results: Circled digit one 8 days after irradiation, the 3H-EG uptake of the irradiation groups was lower than the control (P3H-Tdr uptake of the irradiation groups were lower than the control (P3H-DG uptakes showed no difference (P>0.05), but the 3H-Tdr uptakes of the irradiation groups was still inferior to the control (P3H-Tdr uptakes had no difference (P=0.995). Circled digit two One week after irradiation, the G2-M% and the apoptosis of the external group were above the internal group, but they became no difference after 2 weeks. Circled digit three After irradiation, the spheroid diameters of the internal irradiation group remained unchanged, but the diameters of the external irradiation group were slight larger than the internal ones. The electron microscope observed necrosis and apoptosis in the radiated MTS with the external irradiation group more than the internal ones. Conclusions: Killing effect of the external irradiation is stronger than the same doses of internal irradiation. When perform interstitial treatment using 32P to seek for the best suitable absorption doses, it should not be less than the same doses of external irradiation.
