摘  要:目的 测层粘连蛋白(LN)和纤维粘连蛋白(FN)在成年大鼠视神经再生模型中的表达,探讨坐骨神经、LN和FN在视神经再生中的作用。 方法 选择64只健康成年雄性SD大鼠,随机分成2组,实验组自体视神经-坐骨神经吻合模型32只,对照组自体视神经-视神经吻合模型32只。术后15、30、45、60 d处死动物取吻合神经。利用免疫组化SP法和计算机图像分析,测定LN和FN在各组中的表达。 结果 实验组LN和FN表达水平高于对照组,差别有统计学意义(P<0.05)。 结论 LN和FN涉及成年大鼠视神经再生。坐骨神经移植有助于增加两者表达,促进视神经再生。Objective To investigate the expressions of Laminin (LN) and Fibronectin (FN) in the optic nerve regeneration model of the adult rats. Methods Sixty-four SD rats were randomly divided into two groups. The right optic nerve of all rats was transected. In the experiment group, a 15mm segment of autologuous sciatic nerve was anastomosed to the distal optic nerve stump,and in the control group,the cut ends of optic nerve were re-anastomosed by sutures. Animals were allowed to survive for 15 , 30 ,45 and 60 days. The anastomosed nerves were obtained and expressions of LN and FN were detected by immunohistochemistry and automatic image analysis technique. Results Outgrowth of optic nerve fibres could be seen within the sciatic nerve graft after transplantation. The expression levels of LN and FN were markedly increased in the experiment group with the statistical difference between the two groups(P<0. 05) and little in the control group. Conclusion LN and FN play an important role in the regeneration of the optic nerve. A sciatic nerve graft onto the transected optic nerve is helpful for regeneration of the optic nerve in adult rats.
