摘　　要：目的:探讨抗氧化剂对肿瘤坏死因子(TNF)诱导的U937细胞凋亡的作用.方法:首先用TNF处理U937细胞,提取小片段DNA进行DNA断裂分析;继而以不同抗氧化剂和TNF与U937细胞共同孵育,用台盼蓝拒染法得到细胞存活曲线,并通过RT-PCR、Western blotting检测细胞的bcl-2表达.结果:抗氧化剂可抑制TNF诱导的U937细胞凋亡,上调bcl-2的表达.结论:活性氧中间产物是TNF诱导细胞凋亡的中介体之一.Purpose: To study the biological function of antioxidants in apoptosis induced by tumor necrosis factor (TNF) in U937 cell. Methods:U937 cell were treated by TNF and the low-molecular-weight DNA was abstracted to analysis. Then we in-cubated U937 cell with TNF and different antioxidants,cell viabillity was assayed by Tryban blue exclusion. The bcl-2 expression was also measured through RT-PCR and western blotting. Results:Certain antioxidants could inhibit TNF-induced apoptosis and up-regulate bcl-2 expression in U937 cell. Conclusion: Reactive oxygen intermediates was a mediator of apoptosis induced by TNF.
