解百忧口服液对抑郁模型大鼠行为及中枢神经递质的影响

摘　　要：目的 :研究解百忧口服液对抑郁大鼠行为及中枢神经递质的影响。方法 :选择 Open- Field法评分相近的成年大鼠 ,应用分养和长期不可预见性中等强度应激造成大鼠抑郁模型 ,并观察解百忧口服液对该模型大鼠的治疗作用。定期测定各组大鼠的体重及糖水消耗量 ;以敞箱实验方法测定大鼠水平及垂直活动 ;利用荧光分光光度法测定动物大脑 NE、DA、5- HT和 5- HIAA含量的变化。结果 :与对照组相比 ,抑郁组大鼠体重增加数和糖水消耗量均下降 ;Open- Field法测定行为水平活动和垂直活动次数均下降 ;同时抑郁大鼠脑内 NE、DA、5- HT、5- HIAA含量也明显下降。解百忧口服液治疗组 (5、1 0、1 5g/ kg,ig,连续 1 2 d)能使模型大鼠糖水消耗量增加 ,水平活动和垂直活动次数明显增加 ;同时使模型大鼠脑神经递质 NE、DA、5- HT及 5-HIAA含量明显增加 (P<0 .0 5～ 0 .0 1 )。结论 :本模型为较理想的抑郁动物模型 ,解百忧口服液能明显改善抑郁动物模型的各项指标。Objective To study the effect of Jiebaiyou Oral Liquid on behavior and central monoamine neurotransmitters in depression rats.Methods To select the adult rats which gained the similar scores by open\|field test,the depression model rat was produced by separation and chronic unpredictable mild stress;the therapy effect of Jiebaiyou Oral Liquid was observed in depression model rats.We detected the increased weight,sweet water wastage and crossing score,rearing score of rat by open\|field test at the stated time;determined the content of NE,DA,5-HT and 5-HIAA of animals′ brain by fluorometry.Results The depression model rats′ increased weight and sweet water wastage were lowered,crossing scores and rearing scores were reduced too by open\|field test compared with normal rats;at the same time the content of central neurotransmitters (NE,DA,5-HT and 5-HIAA) of the depression model rats was lowered obviously.Jiebaiyou Oral Liquid (5,10,15g/kg,ig,continuous 12 days)could increase sweet water wastage and the crossing scores and rearing scores of the depression model rats;and Jiebaiyou Oral Liquid could obviously increase the content of central neurotransmitters of the depression model rats.Conclusion This model was the better resembling depression of disease model;Jiebaiyou Oral Liquid could obviously improve all the symptom of the depressive model.
