原发性大肠癌CD44v mRNA表达与DNA含量的关系

摘　　要：目的:探讨原发性大肠癌中CD44v mRNA表达与癌细胞DNA含量之间的关系及其临床意义.方法:应用RT-PCR法检测72例大肠癌中CD44v mRNA的表达情况,并根据CD44v mRNA的表达情况(阴性及微弱表达17例为Ⅰ组,中高度以上表达者55例为Ⅱ组)检测其癌细胞DNA干系水平.结果:72例受检标本中,CD44v mRNA中度以上表达55例(76.39%),其中有淋巴结转移者为93.62%(44/47),无淋巴结转移者为44.00%(11/25).在DNA干系水平方面,Ⅰ组病例二倍体明显高于Ⅱ组病例,异倍体则明显低于Ⅱ组病例(P<0.01).结论:CD44v mRNA的高表达与大肠癌的侵袭转移关系密切,CD44v mRNA表达与大肠癌细胞DNA干系水平的变化一致,再次证实了CD44vPurpose:To study the relationship between the expression of CD44v mRNA and DNA content,and their signifi-cance in the course of metastasis of primary colorectal carcinoma (PCC). Methods:The authors detected the expressions of CD44v mRNA in 72 patients with PCC by RT-PCR. In addition,we determined the DNA stemlines of cancer cell in two group of PCC. Group I (17 cases) expressed CD44v mRNA negatively and weakly. Group n (55 cases) expressed CD44v mRNA highly. Resultsjn 55 of 72 patients with PCC,the expression of CD44v mRNA was moderate or over(76. 39%). Diploid(2C) of DNA of cancer cell in group I was more than that in Group II ,and aneuploid (AN) in group I was lower than that in Group ? (P< 0. 01). The number of patients with lymphatic metastasis was more in group ? than that in Group I . Conclusion:The expres-sion of CD44v mRNA is accordance with DNA stemlines of cancer cell. The results comfirms again that the expression of CD44v mRNA play an important role in the course of PCC metastasis.
