摘 要:目的 :分析Smad 4、Smad 7mRNA和蛋白在人肝癌组织中的表达并与癌旁组织比较 ,了解Smad 4mRNA和蛋白与肝癌临床病理特征之间的关系。方法 :用原位杂交和免疫组化方法检测 36例肝癌存档病理标本和其中的 2 8例癌旁组织病理标本Smad 4、Smad 7mRNA和相应的蛋白。结果 :Smad 4mRNA和其蛋白在肝癌组织中的表达低于肝癌癌旁组织 ,癌组织表达率分别为 2 8%和 19% ,癌旁组织表达率分别为 86 %和 75 % ,两者之间差别有统计学意义(P <0 .0 1)。Smad 7mRNA和其蛋白表达则相反 ,癌组织表达强 ,表达率分别为 6 6 %和 6 4 % ,癌旁组织表达率分别为14 %和 36 % ,两者之间差别有统计学意义 (P <0 .0 1和P <0 .0 5 )。Smad 4mRNA和蛋白与肝癌患者年龄、性别和肿瘤大小无关 ,但与肿瘤细胞类型、是否转移存在相关性。结论 :Smad 4和Smad 7蛋白作为转化生长因子 - β(TGF β)信号转导途径的下游分子 ,在肝癌组织中表达发生了改变 ,参与肝癌的发生、发展。Objective:To study the expressions of Smad 4, Smad 7 mRNA and their proteins in hepatocellular carcinoma, and the relationship between Smad 4 mRNA and its protein expressions and clinicopathologic characteristics. Methods:The mRNA and protein expressions of Smad 4, Smad 7 were detected by situ hybridization and immunohistochemical technique in 36 samples of resected hepatocellular carcinoma, 28 samples of matched non-cancer tissues. Results:The expression rates of Smad 4 mRNA and its protein were significantly lower in hepatic cancer tissues (28% and 19%) than in non-cancer tissues adjacent to cancer (86% and 75%) (P<0.01);The expression rates of Smad 7 mRNA and its protein were differ from those of Smad 4 mRNA and its protein. The hepatic cancer samples (66% and 64% positive) showed a significantly higher level of Smad 7 mRNA and its protein expressions compared with matched non-cancer tissues (14% and 36% positive) (P<0.01,P<0.05).The expressions of Smad 4 mRNA and its protein were related to carcinoma cell types and metastases but were not associated with patient’s age, gender, tumor size. Conclusion:The expressions of Smad 4, Smad 7 mRNA and their proteins in hepatic cancer tissues play an important role in the carcinogenesis and development of hepatic carcinoma.
