摘 要:目的 探讨硫酸化甲壳质对鼻咽癌CNE - 2Z细胞增殖和周期的影响。方法 MTT法检测增值抑制率和IC50 ,流式细胞术分析细胞周期。结果 不同浓度的硫酸化甲壳质分别处理细胞 2 4h、4 8h、72h时 ,抑制率随浓度的增加和时间的延长而增加 ,其IC50 分别为 (0 5 2 9± 0 0 39)、(0 0 4 8± 0 0 2 7)、(0 389± 0 0 2 7)mg/ml,各IC50 间的差异有非常显著性意义 (P <0 0 1)。 0 2mg/ml,0 4mg/ml,0 6mg/ml硫酸化甲壳质分别处理细胞 12h、2 4h、4 8h时 ,G0 /G1期细胞明显下降 ,G2 /M期细胞明显升高 ,随时间的延长变化更明显 (P <0 0 1)。结论 硫酸化甲壳质对CNE - 2Z细胞具有增殖抑制作用 ,并呈剂量和时间依赖性 ;阻滞细胞于G2 /M期 ,也呈时间依赖性 ;一定剂量的硫酸化甲壳质可能通过阻滞CNE - 2Z于G2 /M期而抑制其增殖。Objective To observe the impact of sulfated chitin on cell proliferation and cell cycle in nasopharyngeal cell line CNE-2Z.Methods The inhibited rates of the proliferation and IC 50 values were determined by MTT assay.Cell cycle was analyzed by flow cytometry.Results Cells were treated with sulfated chitin in various concentrations for 24、48、72h,respectively.The increasement of inhibited rates of the proliferation increased with increasement of concentration and incubative time.The IC 50 values at 24、48、72h were (0 529±0 039)、(0 483±0 027)、(0 389±0 027)mg/ml respectively.The difference among IC 50 value was significant (P<0 01).When the cells were treated with sulfated chitin on 0 2,0 4and 0 6mg/ml for 12,24and48h,respectively. the number of the cells in G 0/G 1phase decreased remarkably but increased obviously in G 2/M phase (P<0 01).The longer incubative time was ,the more variation. Conclusion sulfated chitin can inhibit the proliferation of CNE-2Z cells.The inhibition depends on dose and incubative time .Sulfated chitin can arrest CNE-2Z cells in G 2/M phase,it is also in dependence on incubative time.
