摘 要:观察了γ射线、紫外线对白血病细胞株K562中多肽:N?乙酰氨基半乳糖转移酶2(ppGalNAcT2,E.C2.4.1.41)基因mRNA表达的影响,初步探讨ppGalNAcT2与肿瘤发生、发展的相关性。以不同剂量的Coγ60射线、紫外线分别照射K562细胞,继续培养12h后将细胞收集,采用逆转录PCR(RT?PCR)法检测ppGalNAcT2mRNA的表达差异变化。结果显示,γ射线、紫外线照射均可抑制白血病细胞ppGalNAcT2在mRNA水平的表达,提示此两种辐照可能与肿瘤细胞受照射后的浸润和转移性能有关,ppGalNAcT2与此性状间有一定的相关性。To observe the effect of γ-irradiation and UV on the mRNA expressions of ppGalNAcT2 (polypeptide N-acetylgalactosaminyltransferase-2) in K562 leukaemia cells and study the relation of ppGalNAcT2 to tumor’s generating and developing. K562 cells were exposed to different doses of γ-rays and ultraviolet, the expressions at mRNA level of ppGalNAcT2 were examined by reverse transcription polymerase chain reaction (RT-PCR) within 12h. The results showed that the mRNA expressions of ppGalNAcT2 decreased evidently in two irradiated groups, and they were negatively relative to the dosage. The mRNA expressions of ppGalNAcT2 in K562 leukaemia cells could be inhibited by radiation , it is postulated that ppGalNAcT2 might relate to some biological specificity of cancer cells, such as tumor’s invasion and migration.
