摘 要:首先从 8种溶剂中筛选出硫酸作为微波协助提取的浸提溶剂 ,然后用正交试验确立了千层塔生物碱的最佳提取条件。以石杉碱甲和石杉碱乙回收率为指标 ,考察了溶剂倍数、溶剂浓度、微波处理时间等因素。结果表明 ,在室温下微波协助提取的最优条件为 :酸浓度 0 8% (v v) ,液固比例 2 5∶1 ,微波处理时间 90s。 3次重复实验所得石杉碱甲和石杉碱乙回收率分别是 93 7%和 93 9% ,相对标准偏差分别为 1 79%和 1 5 6% (n =3)。与传统的回流提取工艺相比 ,过程时间从 2h缩短为 90s,回收率提高了 1 0 %以上。The process of huperzine A and huperzine B extraction was enhanced with application of microwave field in the aqueous solution. Eight solvents were compared and H 2SO 4 aqueous solution at 1% was selected. An orthogonal design method was used to search for the best experimental condition. The experiment carried out at room temperature with variation of microwave processing time, acid concentration and ratio of the solution to the plant solid. The best result was obtained when the processing time was 90 s, H 2SO 4 concentration 0.8%, and the ratio of the solution to the plant solid 25∶1. Under the optimized condition, the average recoveries of huperzine A and huperzine B were 97.3% and 93.5% respectively. The relative standard deviations for three repeated experiments were 1.79% and 1.56%(n=3). Compared with the traditional method of reflux extraction, the novel extraction method reduced the process time from 2 h down to 90 s with more than 10% increase in the recovery.;
