蛋白酶体抑制剂对雌激素诱发的大鼠催乳素瘤的抑制作用

摘　　要：目的　研究蛋白酶体抑制剂PS - 341对雌激素诱发的大鼠催乳素 (Prolactin ,PRL)瘤的抑制作用。方法　成年雄性SD大鼠 2 5只 ,随机分为 2组 :催乳素瘤组 2 0只 ,皮下植入含有乙烯雌酚的硅胶管 ;对照组 5只 ,皮下植入空白硅胶管。 8周后 ,将催乳素瘤组随机分 3组 ,分别给予蛋白酶体抑制剂PS - 341、诺果宁或安慰剂。治疗 4周后处死大鼠 ,垂体称重 ,做垂体免疫组化检查 ,用放免法测定外周血PRL水平。结果　蛋白酶体抑制剂组和诺果宁组大鼠垂体重量和血清催乳素水平均较安慰剂组明显降低 (P <0 .0 1) ,催乳素阳性细胞计数较安慰剂组明显减少 (P <0 .0 1)。结论　蛋白酶体抑制剂PS - 341对乙烯雌酚诱发的大鼠PRL瘤均具有一定程度的抑制作用。Objective To study the effect of proteasome inhibitor on estrogen-induced rat prolactinoma.Methods Twenty-five adult male SD rats were divided into 2 groups at random.The rats in prolactinoma group (n=20) were subscutaneously implanted with an estrogen-containing implant.The rats in control group(n=5))were implanted with a blank implant. After 8 weeks, rats in prolactinoma group were subdivided into 3 groups proteasome inhibitor group(n=6) received PS-341 0.5 mg/kg twice weekly via tail vein, Norprolac group(n=6) received Norprolac 30 μg/(kg·d) orally, and vehicle group(n=8) received vehicle only. After 4 weeks of treatment, all the animals were sacrificed.Pituitary glands were taken out, weighed,and subjected to immunocytochemical examination. Serum prolactin(PRL) levels were measured by RIA.Results The weights of pituitary glands,PRL immunoreactive cell counts and serum PRL levels in poteasome inhibitor group were lower than those in vehicle group (P<0.01).Conclusion Proteasome inhibitor PS-341 has probable prevetive effect against estrogen-induced rat prolactinoma.
