摘 要:目的:探讨白血病患者TGF-β1及其受体表达在白血病发展过程中的意义。方法:用免疫组织化学或ELISA法检测白血病患者不同病程时期外周血、骨髓以及骨髓基质细胞的结合型TGF-β1,TGFRⅡ和可溶性TGF-β1。结果:初诊患者外周血结合型TGFRⅡ和可溶性TGF-β1含量明显下降,缓解后又升高至接近正常,结合型TGF-β1表达无明显异常。骨髓有核细胞的结合型TGFRⅡ表达情况与外周血基本一致,初诊患者明显下降,缓解后又升高。初诊患者的骨髓基质细胞生长不良,结合型和可溶性TGF-β1含量均明显低下,缓解后又趋于升高,但骨髓基质细胞TGFRⅡ未见明显异常。结论:白血病骨髓基质细胞的TGF-β1表达低下,可能是其外周血和骨髓中TGF-β1含量下降的重要原因,而白血病患者TGF-β1下降及TGFRⅡ表达低下,可能是白血病细胞得以大量增生的重要原因。Objective: To study the significance of transforming growth factor- β1 (TGF- β1) and its receptor, TGFRⅡ expression in different stage of leukemic patients. Methods: Immunohistochemistry or enzyme-linked immunoassay(ELISA) were used to detect soluble TGF- β1 and membrane- binding type TGF-β1 and its receptor, TGFRⅡ in peripheral blood, bone marrow blood and bone marrow stromal cells in different stage of acute leukemic patients. Results: Membrane- binding type TGFRⅡand soluble TGF- β1 in peripheral blood were decrease significantly at first visit. They would increase to normal level after complete remission. Concurrently, the membrane- binding type TGF- β1 was in normal. Membrane- binding type TGFRⅡ on bone marrow nucleared cells were similar to that on peripheral blood nucleared cells, that was a lower membrane- binding type TGF- β1 at first visit and a higher level after complete remission. Stromal cell hypoplasia, decreased membrane- binding type and soluble TGF- β1 were observed in bone marrow stromal cells at first visit. They would increase to normal after complete remission.But the membrane- binding type TGFRⅡ on bone marrow stromal cells was in normal. Conclusion: The reduction of TGF- β1 in bone marrow stromal cells may result to the decreased soluble TGF- β1 in bone marrow and peripheral blood. Low TGF- β1 and TGFRⅡ may result to leukemic cells over proliferation.
