Changes of Phenols Content and Activity of Enzymes Related to Phenols in Lily Bulbs Stored at Different Cold Temperatures for Breaking Dormancy

摘  要:兰州百合鳞茎分别在2℃、6℃、10℃条件下保湿贮藏至101d。贮藏过程中定期取样测定鳞茎各部位酚类物质含量、苯丙氨酸解氨酶(PAL)活性、多酚氧化酶(PPO)活性以及游离苯丙氨酸含量,同时比较了发芽鳞茎与休眠鳞茎中酚类物质含量和PAL活性的差异。结果表明,低温解除休眠过程中,鳞茎内酚类物质含量呈增加趋势。贮藏初期的34d是酚类物质含量以及相关酶活性变化的关键时期。鳞茎不同部位的酚类物质含量呈极显著差异,以鳞茎盘内最高,顶芽次之,鳞片中酚类物质含量较低。顶芽和鳞片的酚类物质含量随贮藏温度升高而下降,且与PAL活性和游离苯丙氨酸含量呈极显著正相关,鳞茎盘中的酚类物质含量变化与之不同。未发现PPO活性与酚类物质含量之间的相关性。Bulbs of Lilium davidii var. unicolor were stored in moist sawdust up to 101 days at 2℃, 6℃or 10℃. The samples of bulbs stored were taken periodically, and were used to determine phenols content, PAL activity, PPO activity and free phenylalanine content in different parts of bulb. Differences in phenols content and PAL activity between dormant and dormancy-broken bulbs were analyzed. The results showed that an increase tendency of phenols content in bulb was observed during the breaking of dormancy. The notable changes in phenols content and activity of enzymes related to phenols occurred in the earlier stage of storage(34 d). Phenols content in different parts of lily bulbs was shown extremely significant differences, and the highest level was in basal plate, and second place was in bud, and lower level was in scales. Phenols content in bud and in scales decreased along with the increase of storage temperature, but that in basal plate showed a different trends. Extremely positive correlations were observed between phenols content and PAL activity or free phenylalanine content. No correlation was found between phenols content and PPO activity.
