摘 要:目的:观察人肝癌裂解抗原致敏的冻融人脐血树突状细胞(dendriticcells,DC)诱导的CTL抗肝癌活性,并观察与细胞因子联用的效应。方法:应用CD34+干细胞试剂盒和免疫磁珠标记法分选人脐血CD34+干细胞,细胞因子扩增培养为成熟DC,液氮冻存;人肝癌裂解抗原致敏冻融DC制备疫苗,进行DC疫苗及其与细胞因子联用的体外抗肿瘤实验,MTT检测各自诱导的CTL活性,并作统计学处理。结果:肝癌抗原致敏的人脐血冻融DC疫苗能诱导高效的CTL活性(P<0.01),与新鲜DC疫苗差异无显著意义(P>0.05)。冻融DC疫苗与1μg/mLGM-CSF联用,或与500U/mLTNF-α联用,诱导的CTL活性均明显增高(P<0.05)。结论:肝癌抗原致敏的人脐血冻融DC疫苗能诱导具有高度针对靶细胞的特异性CTL活性,与细胞因子联用有一定的协同作用。Objective To observe the antitumor effects of CTL primed by tumor cell lysates pulsed frozen dendritic cells (DC) from cord blood and the effects of DC vaccine combined with cytokines. Methods Human cord blood CD34+cells were isolated with CD34+cell kit and MACS. Mature DC were induced from cord blood CD34+cell,and then cryopreserved in liquid nitrogen. DC vaccine was prepared by pulsing frozen DC with human hepatoma cell lysates. The MTT assay was adopted to detect the activity of CTL in the antitumor experiments of DC vaccine and DC vaccine combined with cytokines in vitro. The data were analysed statistically. Results Frozen DC vaccine could induce CTL with potent activity (P< 0.01). There was no significant difference between frozen DC vaccine and fresh DC vaccine (P >0.05). The activity of CTL increased significantly when frozen DC vaccine combined with either 1 μg/mL GM CSF or 500 U/mL TNF α(P< 0.05). Conclusion DC vaccine prepared by pulsing frozen DC derived from human cord blood and pulsed with hepatoma cell lysates could induce CTL with potent activity special to target cells,and have a cooperative effect with cytokines.
