摘  要:应用M13、MYO,LE11.8三种小卫星探针,通过Southern杂交法,检测了8例儿童AML在病程中的DNA指纹变化情况,结果表明:初治或复发时与完全缓解时的DNA指纹不相同,DNA指纹为检测儿童AML的微小残留病灶提供了敏感方法。We detect DNA 'fingerprint' CDNA—F)variation in eight children with acute myeloid leukemia(AML)with MYO.M13.LE11.8 three minisatellite probes by Southern blot hybridization.We found in some patients the DNA—Fs were clearly different at the time of diagnosis and/ or relapse compared with remission,which result from bp increasing or reducing between normal restriction sites.This supports that the leukemia cells of the childhood AMI.have gene rearrang ments.DNA—F do in deed distinguish between normal and leukemia cell clones.In case 3 two DNA—F samples taken during clinical remission were differed,but the latter remission specimen and the subsequent relapse sample were identical.So we think DNA—F can detect minimal residual leukemia cells during clinical complete remission.In case 5 peripheral blood and bone marrow were taken in AML diagnosis.Leukemia cells were 77% in bone marrow,but none was seen in peripheral blood.The DNA—Fs were identical in both samples.So we consider that DNA—F may detect the leukemia cells which are like normal cells in morphology.The southern blot hybridization with DNA—F probes can prove to be a sensitive method in the detetion of minimal residual disease in AML,providing a new way for studying the clonal gene rearrangements of leukemia.
