O型口蹄疫病毒不同基因型联合RT-PCR检测方法的建立

摘　　要：在比较国内外口蹄疫病毒 (FMDV)流行毒株序列的基础上 ,设计了检测 FMDV的 PCR引物及 O型 FMDV不同基因型即中国型 (Cathay)与乏亚株 (Pan Asia)的二联 PCR引物。在确定单项 RT- PCR反应条件的基础上 ,建立、优化了二联 RT- PCR反应体系和条件 ,并进行了相关病毒检测试验。结果表明 ,建立的检测 O型 FMDV不同基因型中国型与泛亚株的二联 RT- PCR,有效、特异且敏感 ,为 FMD的诊断。After PCR primer for FMDV and multiple PCR primers for differention of Cathay and PanAsia topotypes of foot-and-mouth disease type O viruses were designed,the optimal conditions of single RT-PCR and multiple RT-PCR were determined by comparing the products of PCR.The methods of multiple PCR for Cathay and PanAsia topotypes of foot-and-mouth disease type O viruses were proved to be a simple,sensitive,specific technique and this study has paved the way for diagnosis of foot-and-mouth disease and application of vaccines against FMD.
