摘 要:Dmrt基因家族是一个与性别决定相关的基因家族。迄今,已在鱼类、爬行类、鸟类、哺乳类等高等动物中检测到了Dmrt基因的存在。为了进一步探讨该家族在系统进化中的保守性,本研究采用简并PCR技术,扩增了罗氏沼虾(Macrobrachiumrosenbergii)Dmrt基因的DM结构域。经序列分析,获得了Dmrt基因家族的3个成员,其编码序列分别与人DMRT2、DMRT3、DMRT4基因DM结构域编码序列的相似性分别为93%、80%和95%,根据罗氏沼虾的拉丁名分别命名为MrDmrt2、MrDmrt3、MrDmrt4。与其他动物相关的Dmrt基因进行聚类分析,结果表明,不同进化地位动物的Dmrt基因DM域编码序列存在高度的同源性,显示Dmrt基因在系统进化上高度保守,序列上的相似性可能暗示着它们在功能上的保守性。The Dmrt genes constitute a new gene family related to sex-determination.Like the double-sex gene of Drosophila melanogaster and the Mab-3 gene of Caenorhabditis elegents,they encode transcription factors characterized by a conserved zinc-finger like DNA-binding motif,the DM domain ,which is thought to bind DNA in the process of sex differentiation and development.In 1998,Dmrt1 genes were first identified and found containing DM domains in Homo sapiens and Mus musculus.And Dmrt1 was found to regulate sex determination and differentiation in vertebrates extensively.So far,the Dmrt genes have been discovered in a wide range of animal species,such as fish,amphibian,reptiles,birds,and mammals.These evidently revealthe evolutionary conservation of Dmrt gene family.But the research of Dmrt genes in crustacean,which was a very important group in phylogenic evolution,has not been reported.Macrobrachium rosenbergii,which is subjected to Macrobrachium,Palaemonidae,Crustacea,is a very important kind of freshwater edible shrimps in China and the economic value is prominent.As to Macrobrachium rosenbergii,sex was an important economic character,for the male are bigger than the female .So to clarify the mechanism of Macrobrachium rosenbergii sex determination will be of great importance in both theory and application.With the hope of doing further research on the functions of Dmrt genes and clarify the molecular mechanisim of Macrobrachium rosenbergii,degenerate PCR was used in this research to amplify the conserved DM domains of Dmrt genes in Macrobrachium rosenbergii.The PCR products were approximately 150bp.After DNA cloning SSCP analysis and sequencing,three different DM sequences were identified.The amino acid sequence of one clone is 93% identical with human DMRT2,named MrDmrt2,according to its name Macrobrachium rosenbergii.Another sequence encodes an amino acid sequence with 80% identical with human DMRT3,named MrDmrt3.The other sequence encodes an amino acid sequence with 95
