摘  要:激光扫描细胞仪(Laser Scanming Cytometer,LSC)是kamentsky等开发的一种新的细胞分析仪,它同时具有流式细胞仪和图像细胞仪的功能。恶性肿瘤自身内在的基因不稳定性可以导致肿瘤发展过程中基因异常的增加,基因不稳定性可分为两类:微卫星不稳定性和染色体不稳定性,目前研究已经证实染色体不稳定性与DNA非二倍体这间有密切关系。在本研究中,我们主要观察肿瘤恶性程度与LSC分析结果的相关性。方法:应用LSC分析脑肿瘤细胞的DNA指数(DI)和增殖指数(PI),使用流式细胞仪(FCM)对脑肿瘤细胞系进行DNA分析,同时对肿瘤标本进行冰冻切片病理检查,与LSC的分析结果进行比较。结果:DNA指数和增殖指数反映了肿瘤的恶性程度。结论:应用LSC进行脑肿瘤DNA分析结合病理学检查对决定术中胶质瘤切除面积是非常有意义的,具有一定的临床应用价值。BACKGROUND & OBJECTIVE: A novel type of cytometry, Laser Scanning Cytometry(LSC) developed by Kamentsky has advantages of both flow and image cytometry. Malignant tumors show intrinsic genetic instability, resulting in an increase in genetic abnormalities with tumor progression. Genetic instability is devided into two types, microsatellite instability (MIN) and chromosomal instability (GIN), and a close link be-tween GIN and DNA aneuploidy in malignant tumors has been demonstrated. In this article, We observed the correla-tion between the malignancy of various brain tumors and the results of DNA -ploidy by LSC. METHODS: The DNA-index (DI) and proliferating index (PI) of the brain tumors were determined by LSC and Flow Cytometer(FCM), and the specimens were also given pathological examination at the same time. The results were com-pared. RESULTS: The DI and PI reflect the malignancy of the brain tumors. CONCLUSION: The analysis of brain tumor DNA by LSC was useful to decide the area that should be resected for glioma in the operation and was worth applying clinically.
