摘 要:目的 :深入研究高浓度氟对神经细胞磷脂脂肪酸组成的影响。方法 :体外培养SH SY5Y人脑神经母细胞瘤细胞 ,在培养液中加入不同浓度的氟化物 ,培养 4 8h后用过柱法分离细胞生物膜中磷脂部分 ,气相色谱法测定磷脂分子中脂肪酸含量和组成 ,比色法测定共扼二烯含量。结果 :经高浓度氟 (5mmol/L及 0 5mmol/L)处理的神经细胞磷脂分子中脂肪酸组成发生改变 ,表现为多不饱和脂肪酸组成减少 ,饱和脂肪酸比例增加 ,减少的多不饱和脂肪酸种类以花生四烯酸和二十二碳六烯酸为主 ;脂质过氧化产物共扼二烯含量增加。结论 :高浓度氟处理SH SY5Y神经母细胞瘤细胞后可引起细胞生物膜磷脂分子中脂肪酸组成发生异常改变 ,其机制与脂质过氧化水平升高有关。Objective: To investigate the influence of fluoride on fatty acid composition of phospholipid in SH SY5Y cells. Methods: SH SY5Y cells, a human neuroblastoma cell line, were incubated with different concentrations of fluoride for 48 hr. Phospholipid was separated by Silica SepPak collumn and fatty acids were measured by Gas Chromatography. Level of conjugated dienes in the cells was detected by a method connected with chromatography. Results: In the groups treated with high-doses of fluoride (5 mmol/L and 0 5 mmol/L) as compared to control, the modification of fatty acid composition was obtained, in which the proportion of polyunsaturated fatty acids was decreased whereas saturated fatty acid increased.
