摘  要:目的探讨神经性钙粘附分子(N-cadherin)在癫痫状态后海马苔藓纤维出芽和突触重组中的作用。方法取锂-匹罗卡品诱导大鼠癫痫持续状态及慢性自发性颞叶癫痫发作期的大鼠脑片,用Timm染色和免疫组化的方法分别检测苔藓纤维出芽和N-cadherin在大鼠海马组织中的表达。结果癫痫状态后第2周和第4周的实验组大鼠可见到苔藓纤维出芽,穿越齿状回颗粒细胞层到达内分子层,并在此形成一条致密的层状带(Timm染色)。免疫组化染色发现实验组大鼠在第2周和第4周,海马齿状回内分子层可以看到强染色,并形成一条致密带,与Timm染色时观察到的条带一致。结论癫痫状态后在海马齿状回内分子层N-cadherin的表达上调,N-cadherin可能参与了癫痫后苔藓纤维出芽和突触重组过程。Objective To explore the correlationship between neural calcium-dependent cell surface adhesion molecule(N-cadherin) and the process of aberrant mossy fiber sprouting and synaptic plasticity following status epilepticus.Methods By Timm and immunohistochemical staining,the process of aberrant mossy fiber sprouting and the expression of N-cadherin was observed in the Lithium-pilocarpine model of epilepsy characterized with an acute period of status epilepticus followed by spontaneous recurrent seizures temporal lobe epilepsy.Results Pilocarpine-induced status epilepticus induced intense Timm's staining in the inner molecular layer of the dentate gyrus at 2 and 4 weeks after seizure.N-cadherin immunostaining showed an increase of N-cadherin in the inner molecular dentate gyrus after pilocarpine treatment,a pattern that was watched identically with the Timm's staining.~Conclusions N-cadherin may be involved in the process of mossy fiber axonal outgrowth and synaptogenesis in the hippocampal dentate gyrus.
