摘 要:目的观察组代谢型谷氨酸受体阻断剂α-methyl-(4-tetrazolyl-phenyl)glycine(MTPG)对脑缺血预处理(cerebralischemicpreconditioning,CIP)抗损伤性缺血所致海马神经元凋亡作用的影响。方法24只SD大鼠采用四血管闭塞法制造大鼠全脑缺血模型。采用Tunel法检测海马CA1区锥体细胞凋亡情况。结果假手术组几乎无阳性细胞表达;损伤性缺血组可见明显的阳性细胞表达和核固缩,阳性细胞数、总面积、平均光密度明显增加(P<0.05);CIP+损伤性缺血组阳性染色细胞数、总面积、平均光密度较损伤性缺血组明显降低(P<0.05);MTPG+CIP+损伤性缺血组阳性染色细胞数、总面积、平均光密度与CIP+损伤性缺血组相比明显增加(P<0.05)。结论mGluR2/3通过抑制凋亡,参与BIT的诱导。Objective We observed effects of cerebral ischemic preconditioning (CIP) on apoptosis induced by lethal ischemic insult,and influence of α-methyl-(4-tetrazolyl-phenyl) glycine(MTPG),the antagonist of metabotropic glutamate receptor 2/3 (mGluR2/3),on the effect of the CIP.Methods By using 4-vessel occlusion to make the global ischemic moded in twenty-four SD rats.Apoptosis of pyramidal neuron in the CA_1 subfield of hippocampus was examined using labeling method.Resuls TUNEL staining showed that there was almost no positive expression in hippocampal CA_1 subfield in sham group.Obviously expression characterized with intensive staining in the cytoplasm and karyopyknosis was found in the ischemic insult group.The number,total area and average light density of the positive cells were significantly increased compared with sham group (P<0.05).While compared with ischemic insult group,the number,total area and average light density of the positive cells in the CIP+ischemic insult group were decreased significantly.In the MTPG+CIP+ischemic insult group,the number,total area and average light density of the positive cells were significantly increased compared with those of the CIP+ischemic insult group (P<0.05).Conclusion mGluR2/3 might participate in the induction of BIT by inhibiting apoptosis.
