摘 要: 目的:探讨他莫昔芬(TAM)对人胰腺癌细胞增殖的影响及其机制。方法:采用钙离子荧光指示剂flour3/Am,流式细胞仪动态检测TAM作用后人胰腺癌细胞(CapanⅡ)内游离钙离子([Ca2+]i)水平的变化,同时用MTT比色法测定细胞增殖情况。结果:TAM使细胞内[Ca2+]i水平上升,与静息状态细胞内[Ca2+]i水平相比差异有统计学意义,P<005。高浓度的TAM对癌细胞的增殖有抑制作用。结论:TAM有抑制人胰腺癌细胞增殖的作用,其作用机制除通过竞争性结合雌激素受体来抑制雌激素的作用外,还可通过诱导细胞内[Ca2+]i水平升高,抑制Ca2+依赖蛋白激酶C和钙调蛋白,从而抑制癌细胞的生长。OBJECTIVE: To study the effect and the mechanism of tamoxifen(TAM) on the proliferation of human pancreatic cancer cell lines.METHODS:Flow cytometry was used to continuously monitor the change of the intracellular free calcium [Ca 2+]i with calcium fluoresence indicator (fluo-3/Am),meanwhile the MTT colorimetry was used to detect proliferation of human pancreatic cancer cell lines(Capan-Ⅱ).RESULTS:TAM made the intracellular[Ca 2+]i concentrations maintained a higher level and was much higher than that of the intracellulay [Ca 2+]i of static status cells, P<0.05. TAM had the effect on inhibiting the proliferation of Capan-Ⅱ cells.CONCLUSION:TAM can inhibit proliferation of human pancreatic cancer cells. The mechanisms of TAM are that it can competitively combine the estrogen receptor and increase the intracellular calcium level to inhibit the activity of the Ca 2+-dependent protein kinase C and calmodulin and inhibit proliferation of cancer cells finally.
