摘  要:采用20个随机引物对洞庭湖区三种不同体色的黄鳝群体(分别以A、B、C区分)进行随机扩增多态性DNA分析。结果12个引物的扩增效果良好,统计学分析表明,黄鳝A与黄鳝B的遗传距离较小(0 054107)且与A、B两个群体内的遗传距离(分别为0 045000、0 038217)相近,说明黄鳝A、B两个群体的遗传差异不明显;而黄鳝C与黄鳝A的遗传距离(0 123654)以及黄鳝C与黄鳝B的遗传距离(0 134074)均明显大于黄鳝A与黄鳝B的遗传距离,且由引物S326和S462扩增出的5条特异性谱带可以作为区分黄鳝C与黄鳝A、B两个群体的遗传标记,说明黄鳝C与黄鳝A、B两个群体之间遗传差异性十分明显。The genotypes of Monopterus albus populations with three colors (population A, B and C) were analyzed in Dongting Lake by random amplified polymorphic DNA(RAPD in which twenty 10-nucleotide-long random primers with 12 well-amplified and reproducible band patterns were used. The results showed that the average genetic distances(0.054107)was lower between population A and B, which was close to the interpopulation average genetic distance of the two population(0.045000 and 0.038217, respectively). This implied that there is no significant difference in genetic differentiation between population A and B. Between population C and A(0.123654)and between population C and B(0.134074), however, the average genetic distances were significantly higher than that between population A and B. Two primers(S326, S462)produced five banding patterns that could be used to discriminate between the population C and the population A and B, suggesting that the genetic differentiation between the population C and the population A and B was remarkable.
