摘 要:目的 观察缺氧复合烧伤血清对心肌细胞NF κB的影响以及在此条件下高牵张刺激对心肌细胞NF κB的影响。方法 采用原代培养的乳鼠心肌细胞 ,分别给予正常血清 (SN组 )、烧伤血清 +缺氧 (1%O2 ) (SH组 )、烧伤血清 +缺氧(1%O2 ) + 10 %轴向静态牵张 (SHS组 ) 3种处理 ,采用Westernbloting检测p5 0、p65、IκB α表达水平 ,并以免疫荧光检测p5 0核转位情况。结果 同SN组比较 ,SH组、SHS组p5 0、p65表达增强 ,SHS组p5 0、p65表达水平较SH组升高 ,p5 0发生较明显的核转位而IκB α表达水平却下降。结论 缺氧复合烧伤血清促进心肌细胞NF κB的表达与核转位 ,机械牵张加强了这一作用。Objective To observe the effects of high stretch on NF-κB in cardiomyocytes under burn sera combined with hypoxia. Methods Ventricular cardiomyocytes isolated from neonatal rats were employed in this study. The cells were stimulated by sera of normal rats (SN group), burn sera and hypoxia (SH group), burn sera and hypoxia and 10% axial static stretch (SHS group), respectively. Expressions of p50, p65, and IκB-αin the cardiomyocytes were determined by Western blotting and the translocation of the former two was determined by immunofluorescence. Results Enhanced expression and activation of p50 and p65 were found in SH and SHS groups as compared with those in SN group. The expression and activation of p50 and p65 in SHS group were higher than those in SH group, but IκB-α decreased at the same time. Conclusion Mechanical stretch can enhance the expression and nuclear translocation of NF-κB resulted from hypoxia combined with burn sera.
