摘  要:目的 观察氟硝丁酰胺对喉鳞癌细胞株HEP 2的生长调控作用。方法 应用MTT比色法、平板集落形成法、流式细胞术检测氟硝丁酰胺对喉鳞癌细胞生长及增殖周期的影响。同时 ,用电子显微镜检查细胞形态学的改变。结果 经氟硝丁酰胺作用后 ,喉鳞癌细胞株HEP 2增殖明显受到影响。与对照组比较 ,P <0 .0 5。细胞抑制率达到 92 .0 8% ,且有明显时间和剂量依赖性 ;流式细胞仪检测细胞增殖周期有滞于G0 /G1期的趋势。并可见凋亡小体形成。结论 氟硝丁酰胺对喉鳞癌细胞具有抑制生长和诱导凋亡的作用 ,为喉癌的性激素临床治疗提供了理论依据。Objective To investigate the effect of flutamide on laryngeal carcinoma cell lines HEP-2 apoptosis.Methods MTT assay, colony formation test and flow cytometer sense were used to measure the proliferation inhibition of laryngeal carcinoma cell lines HEP-2.At the same time, the morphology of tumor cells were checked with electron microscope. Results Flutamide inhibited laryngeal carcinoma cell lines HEP-2 obviously with dose and time dependence. The inhibition rate was 92.08%. The statistical difference between flutamide group with anhydrous alcohol group was very significant(P<0.05). The result of flow cytometry showed the tendency that flutamide could be inhibit the process of cell cycle from G 0 to G 1 stage. Apoptotic body can observed with electron microscope.Conclusion Flutamide induced the apoptosis, inhibited the growth of laryngeal carcinoma cell lines HEP-2. Further study is needed to elucidate the role of antiandrogen therapy in laryngeal carcinoma cell.
