摘 要:利用紫外线和亚硝酸对黑曲霉进行联合诱变,使用自制的培养基筛选出7株对羊毛细化具有效果的菌株,又通过摇瓶复筛出1株对羊毛细化具有明显效果的菌株。同时设计了1种以羊毛作为底物的酶活力的测定方法,用这种测定方法,筛选出的菌株的最高产酶活力可以达到620μg ml。A high yield acid protease producing strain Asp.niger DH-C40 is screened from its parent strain Asp.niger DH-010 by treating with UV and nitrous acid. The acid protease is good at decompose wool. Invent a new way to determine the activity of the enzyme for wool. Define 1 ml enzyme decompose 1 μg wool in one hour as 1 unit activity of enzyme. A new screening culture medium was designed. Using the new culture medium, 7 strains which can produce the enzyme for wool are screened. Using the flask bottle to screen among the 7 strains, we get a high yield strain——DH-C40. Using the new determine way, DH-40 can produce a high yield acid protease for wool, the activity of protease is 620 μg/ml.
