摘　　要：目的　探讨高强度超声治疗对患宫颈癌 U14小鼠脾淋巴细胞增殖及细胞毒性影响。方法　采用四唑盐比色法 (MTT法 )及双色流式细胞技术检测了高强度超声 (HIU)治疗组 ,手术治疗组 ,患瘤对照组及正常对照组小鼠脾淋巴细胞的增殖功能及细胞毒性。结果　HIU治疗组小鼠脾淋巴细胞增殖活性较手术组、患瘤对照组及正常对照组增高 (P<0 .0 5 ) ,手术组及正常对照组较患瘤对照组小鼠脾淋巴细胞增殖活性增强 (P<0 .0 5 ) ,HIU治疗组脾淋巴细胞对同源宫颈癌 U14细胞有较强的杀伤活性 ,与手术、患瘤及正常对照组比较差异明显 (P<0 .0 5 )。结论　HIU治疗患宫颈癌 U14小鼠后 ,原位埋植的固化肿瘤细胞具有同源肿瘤的抗原性 ,能够激活小鼠脾淋巴细胞 ,发挥抗肿瘤免疫功能 ,起到“HIU原位固化瘤疫苗”Objective To investigate the effect of HIU therapy on the splenic lymphocyte proliferation and splenic lymphocyte cytotoxicity in BALB/c mice bearing U14 cervical cancer.Methods All 32 mice were divided into 4 groups:HIU therapy group,surgery resection group,no-treatment group and normal group.Splenic lymphocyte proliferation and its lymphocyte cytotoxicity were determined using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide( MTT )and double-colour flow cytometry respectively.Results The A570 values in HIU group,surgery resection group,no-treatment group and normal group were:0.412±0.031,0.247±0.055,0.136±0.011 and 0.263±0.027 respectively.The A570 value in group A was higher than that in group B,C and D,P<0.05.When effective cells /targeted cells=5:1,the death rates in group A,B,C,D were 55.38±8.16%,43.91±5.69%,31.20±7.12%,32.28±5.84% respectively and the death rate in group A was higher than that in group B,C,D(P<0.05).When effective cells /target cells=10:1,the death rates in group A,B,C,D were 56.72±6.40%,45.28±6.47%,37.33±7.68% and 39.85±8.59%,respectively;the death rate in group A was also higher than that in group B,C,D(P<0.05).Conclusions We think that the U14 cervical carcinoma cells solidified by HIU may have the homologous antigen as U14 cervical carcinoma cells,and the solidified carcinoma cells can stimulate splenic lymphocyte to accelerate its activation and proliferation.Therefore,the solidified carcinoma cells can act as HIU solidified vaccine.
