摘 要:目的观察活血化瘀中药的有效成份川芎嗪和苦参碱对HSC-T6细胞增殖的作用。方法经传代培养,绘制肝星状细胞HSC-T6的细胞生长曲线,并计算其细胞群体倍增时间和克隆形成率等基本的细胞生物学参数;应用MTT比色法测定HSC-T6细胞增殖及评价药物对细胞增殖的作用。结果HSC-T6细胞成活率较高,细胞在指数增殖期的细胞群体倍增时间为10.57h,克隆形成率为82.4%。川芎嗪在10-9~10-4mol/L浓度范围之内,抑制HSC细胞增殖作用呈剂量依赖性,与对照组相比均具有显著性差异(P<0.05)。苦参碱在10-8~10-4mol/L浓度范围之内,抑制HSC细胞增殖作用呈剂量依赖性,与对照组相比均具有显著性差异(P<0.05)。结论川芎嗪和苦参碱对HSC-T6细胞增殖具有抑制作用,呈剂量依赖性。Objective To investigate the effects of active materials, chuanxingzine and matrine, on the proliferation of HSC T6 cells. Methods Growth curve of HSC T6 was drawn and some basic indicators of HSC T6 cell biology were calculated, such as population doubling time and cloning efficiency. The anti proliferation effects of the drugs on HSC T6 cell were evaluated by MTT assay. Results The population doubling time of HSC T6 was 10.57 hours and its cloning efficiency was 82.4%. Ranging from 10-9mol/L to 10-4mol/L, chuanxingzine had the dose dependent inhibitory effect on the proliferation of HSC T6 cells, which showed significant difference compared with control group(P< 0 05). Ranging from 10-8mol/L to 10-4mol/L, matrine also had the dose dependent inhibitory effect on the proliferation of HSC T6 cells, which showed significant difference compared with control group(P < 0 05). Conclusion Chuanxingzine and matrine have the dose dependent inhibitory effect on the proliferation of HSC T6 cells.
