摘  要:目的 检测环氧合酶 -2 (cyclooxygenase ,COX -2 )在人晶状体上皮细胞 (humanlensepitheliumcell,HLEC)中的表达 ,探讨COX -2在后发性白内障 (PCO)形成中的意义。方法 细胞传代培养 ,取第三代细胞用于实验。用SP免疫组化法检测bFGF刺激 6小时后COX -2蛋白的表达 ,并与对照组相比较 ,以胞浆内出现棕黄色颗粒为阳性表达 ;采用RT -PCR和Western -blot方法检测bFGF刺激下 ,不同时间点 (1h、 3h、 6h、 12h、 2 4h)COX -2的mRNA和蛋白的表达。结果 检测的人晶状体上皮细胞中 ,实验组的细胞COX -2mRNA和蛋白表达均为阳性 ,且表达呈时间依赖性 ,随着时间的延长COX -2表达逐渐增加 ,6h达最高峰 ;正常对照组表达弱或无表达。结论 bFGF可以刺激晶状体上皮细胞增殖 ,并诱导环氧合酶 -2的表达 ,提示COX -2与人晶状体上皮细胞的增殖密切相关。COX -2可能参与PCO的形成。Objective To measure the expression of cyclooxygenase-2(COX-2)in human lens epithelial cell(HLEC)and to explore the significance of COX-2 in the formation of posterior capsule opacification(PCO).Methods Cell was subculture and the third-generation cell was used for the experiment.The expression of COX-2 protein stimulated by bFGF for 6 hours was measured by SP immunohistochemistry and brown granula were positively expressed in cytoplasm in comparison with the control group.The expression of COX-2 mRNA and protein stimulated by bFGF were measured by RT-PCR and Western-blot at different time periods.Results Cell in test group expressed COX-2 and protein positively,with the characteristic of time dependence.The expression of COX-2 became more significantly as time prolonged,hitting the peak after 6 hours.By comparison,the expression in control group was slight or negative.Conclusions bFGF can induce the expression of COX-2 in human lens epithelial cells;COX-2 has great relation with the proliferation of human lens epithelial cells.Therefore,COX-2 may be involved in the formation of PCO.
