摘 要:目的 研究重组人生长激素 (rhGH)在体内外对胃癌细胞生长的影响。方法 将大鼠分为对照组、rhGH组、奥沙利铂 (L OHP)组和rhGH +L OHP组 4组 ,利用流式细胞仪和裸鼠移植瘤实验等方法测定rhGH对人胃癌细胞株BGC82 3细胞周期、增殖指数 (PI)、肿瘤抑制率和移植瘤核增殖抗原(PCNA)的影响。结果 rhGH组G0 ~G1 期细胞数占 5 2 3% ,S期细胞数占 32 6 % ,G2 ~M期细胞数占14 7% ,PI为 4 7 7% ,肿瘤抑制率为 5 % ,PCNA阳性表达率为 82 7% ,与对照组 (G0 ~G1 期细胞数占4 7 8% ,S期细胞数占 37 0 % ,G2 ~M期细胞数占 15 3% ,PI 5 2 3% ,肿瘤抑制率 0 ,PCNA77 8% )相比差异无显著意义 ;rhGH +L OHP组 (G0 ~G1 期细胞数占 86 5 % ,S期细胞数占 5 5 % ,G2 ~M期细胞数占 7 8% ,PI13 5 % ,肿瘤抑制率 4 9 3% ,PCNA 5 8 2 % )与L OHP组 (G0 ~G1 期细胞数占 83 9% ,S期细胞数占 7 9% ,G2 ~M期细胞数占 8 3% ,PI16 2 % ,肿瘤抑制率 4 6 6 % ,PCNA 6 2 5 % )比较差异亦无显著意义 ;rhGH +L OHP组与rhGH组和对照组比较 ,G0 ~G1 期细胞肿瘤抑制率明显增加 ,S期细胞、PI和PCNA明显减少。结论 重组人生长激素在体内外不会促进胃癌细胞的增殖。Objective To study effects of recombinant human hormone (rhGH) on the growth of a human gastric carcinoma cell line in vitro and in vivo Methods The experiment was divided into control group, rhGH group, oxaliplatin (L OHP) group and rhGH+L OHP group. Cell cycle, cell proliferation index (PI), tumor inhibitory rate and the expression of proliferating cell nuclear antigen (PCNA) of human gastric cancer cell line (BGC823) were examined by flow cytometry and nude mouse model Results There was no statistical difference between rhGH group (G 0~G 1 phase 52 3%, S phase 32 6%, G 2~M phase 14 7%,PI 47 7%, the tumor inhibitory rate 5%, PCNA 82 7%) and control group (G 0~G 1 phase 47 8%, S phase 37 0%, G 2~M phase 15 3%, PI 52 0%, the tumor inhibitory rate 0%, PCNA 77 8%) or between rhGH+L OHP group (G 0~G 1 phase 86 5%, S phase 5 5%, G 2~M phase 7 8%, PI 13 5%, the tumor inhibitory rate 49 3%, PCNA 58 2%) and L OHP group (G 0~G 1 phase 83 9%, S phase 7 9%, G 2~M phase 8 3%, PI 16 2%, the tumor inhibitory rate 46 6%, PCNA 62 5%) ( P >0 05)in all parameters The tumor inhibitory rate and the cells detained in G 0~G 1 phase significantly increased, meanwhile, cells in S phase, the PI and PCNA significantly decreased in rhGH+L OHP group compared with control group or rhGH group ( P <0 01) Conclusion In vivo and in vitro rhGH does not accelerate the proliferation of human gastric cancer cells
