摘  要:目的 探讨体外内毒素 (LPS)对LO2 细胞的直接作用以及N 乙酰半胱氨酸 (N acetylcysteine,NAC)的抑制效果。方法 将LO2 细胞培养 2 4h后换用含LPS(2 0 μg/ml)和 /或NAC(5mmol/L)的新鲜培养液 ,提取肝细胞的基因组DNA ,经琼脂糖凝胶电泳观察肝细胞生物化学性质的改变 ;用透射电镜研究其形态学变化 ;TUNEL标记法检测细胞凋亡。结果 LPS直接作用的LO2 细胞在形态学上表现出胞浆浓缩核凝聚成团块、形成凋亡小体、细胞器结构基本正常等凋亡细胞的特征。将NAC与LPS同时加入培养液 ,作用 16h发现NAC可明显减少因LPS所诱导的LO2 细胞的凋亡。结论 LPS在体外可诱导肝细胞凋亡 ,NAC能抑制其作用。Objectives To investigate the effects of lipopolysaccharide(LPS) and N-acetylcysteine(NAC) on LO 2 cells in vitro.Medthods The LO 2 cells were cultured for 24h in vitro before the fresh culturing medium with LPS(20μg/ml) or/and NAC were added. DNA-fragment assay was analyzed by the agarose gel electrophoresis to determine apoptostic level. Transmission electron microscopy techniques were used to observe the morphological changes of hepatocyte treated with LPS. The apoptosis of LO 2 cells detected by terminal deoxynucleotide transferas-mediated aUTP nick end labeling(TUNEL). To investigate the restrainable effects of N-acetylcysteine(NAC) .Results LO 2 cells were treated with LPS exhibited some typical apoptosis-specific morphological features. The LPS-induced apoptosis cells decreased when the culture medium contained with LPS and NAC were added for 16h.Conclusions NAC can restrain LPS-induced apoptosis of hepatocyte in vitro.
