Establishment and biological characteristics of choriocarcinoma in nude mice

摘  要:目的通过建立绒癌JAR细胞的裸小鼠动物模型,探讨有关其生物学特征。方法将JAR细胞接种至裸小鼠胁腹部皮下,动态观察肿瘤生长情况和带瘤生存时间;尸解裸小鼠,取组织行显微镜下观察和作透射电镜检查;监测血hCG变化(FIA);免疫组化SP法检测肿瘤组织的hCG和PECAM1表达情况。结果JAR细胞皮下移植瘤的成功率为90%,移植瘤呈紫蓝色,切面出血坏死明显;显微镜下见滋养细胞大量增生,有大量血窦和新生血管形成;电镜下滋养细胞核分裂相多见,核仁多个,胞浆富含线粒体和内质网。免疫组化染色示滋养细胞hCG(+)、PECAM1(-),血管内皮细胞hCG(+)、PECAM1(+)。结论与正常妊娠过程中滋养细胞侵袭母体血管机制相似,恶性滋养细胞也能够侵犯裸小鼠的血管,并取代宿主的血管内皮细胞;hCG可能与血管生成有关。Objective Establishing an animal model using JAR choriocarcinoma cells grown in nude mice, we investigated the biological characteristics and applications of it. Methods We injected JAR cells into the subcutaneous space of the flank of nude mice,observing the growth of choriocarcinoma and the survival time of them with tumor. After autopsy,we inspected the tumors under light microscope and electron microscope. Meanwhile we determined levels of hCG in serum of nude mice with FIA(fluoroimmunometric assay). Expressions of hCG and PECAM-1 in tumor tissue were detected by immunohistochemistry(SP). Results Ninety percent nude mice inoculated with JAR cell line showed the growth of choriocarcinoma. JAR choriocarcinoma were bluish in color and bled heavily after section with trophoblast cell proliferating prominently. A large number of lacunae-like space filled with blood and neovascularization could be observed under light microscope. The tumor had high mitotic activity with multiple nucleoli under electron microscope. Mitochondria and endoplastic reticuli were abundant in cytoplasm. HCG was positive in tumor cells immunohistochemically as well as hCG and PECAM-1 positive in endothelial cells of blood vessels,while PECAM-1 was negative in tumor cells. Conclusion JAR choriocarcinoma cells invaded host vessels and replaced the endothelial lining in a similar way to trophoblast cells during the establishment of a haemochorial placenta. HCG may have effect on angiogenesis. This subcutaneous xenograft of JAR choriocarcinoma cells can be useful in investigating clinical test of drugs.
