摘  要:目的 观察大鼠局灶性脑缺血 /再灌注后脑梗死体积的变化和葡萄糖转运体 1(GLUT1)在缺血半影区的表达情况以及人参皂甙Rb1对其的影响。方法 雄性成年SD大鼠 42只 ,随机分成 3组 :假手术组、对照组和人参皂甙Rb1干预组。用线栓法复制大脑中动脉阻塞 (MA CO)模型 ,在脑缺血 1h再灌注 5h时取材 ,用氯化三苯基四唑氮 (TTC)染色后 ,全自动图像分析系统进行图像分析脑梗死体积比 ;逆转录 聚合酶链反应 (RT PCR)测定GLUT1mRNA水平的变化 ;用免疫组织化学半定量测定GLUT1蛋白水平的变化。结果 假手术组、对照组和人参皂甙Rb1干预组的梗死体积百分比分别为 0、(0 .0 4± 0 .0 1) %和 (0 .0 2± 0 .0 1) % ;3组半影区的GLUT1mR NA水平分别是 (0 .2 3± 0 .14 )、(0 .5 5± 0 .0 4)和 (0 .79± 0 .19) ;蛋白表达的PU值分别为 (5 .1±1.2 )、(13 .1± 1.7)和 (19.0± 1.9)。结论 人参皂甙Rb1明显缩小缺血再灌注损伤后脑梗死的体积 ,Rb1通过上调半影区GLUT1表达以维持脑组织的能量供给可能是其发挥脑保护作用的机制之一。Objective To investigate the change of infarct size and the expression of GLUT1 in penumbra area after cerebral ischemia/reperfusion injury in rats and the effect of ginsenoside Rb1 on them.Methods Forty-two male adult SD rats were randomly divided into 3 groups:sham-operated group,the control group and the ginsenoside Rb1 group.The model of middle artery occlusion (MCAO) in rats was established by using an intraluminal thread technique.The influence of ginsenoside Rb1 on infarct size,neurologic deficit,the expression level of GLUT1 mRNA and protein in the penumbra were observed.The infarct size was analyzed quantitatively by Kontron IBAS 2.5 image auto-analyses system.The change of GLUT1 mRNA was assessed by RT-PCR.The expression of GLUT1 protein was detected by immunohistochemistry.Results The volume percentage of infarct in sham-operated group,the control group and the ginsenoside Rb1 group was 0, (0.04± 0.01) % and (0.02± 0.01) % respectively (P<0.01).The level of GLUT1 mRNA in sham-operated group,the control group and the ginsenoside Rb1 group was 0.23± 0.14, 0.55± 0.04 and 0.79± 0.19 (P<0.05) and the PU value of GLUT1 protein expression was 5.1± 1.2, 13.1± 1.7 and 19.0± 1.9 respectively (P< 0.05).The GLUT1 mRNA and protein levels in ginsenoside Rb1 group were much higher than in the control group.Conclusion Ginsenoside Rb1 could decrease the infarct size after cerebral ischemia/reperfusion injury.Rb1 maintained the energy supply of the brain via upregulating GLUT1 expression,which may be one of the protective mechanisms.
