摘  要:目的 研究培高利特(pergolide)和螺哌隆(spiperone)对沙土鼠前脑缺血再灌注期纹状体多巴胺 (DA)代谢的影响。方法采用双侧颈总动脉阻断法制备沙土鼠(64只)前脑缺血再灌注损伤模型,缺血5分钟。 将实验动物分为四组。假手术(Sham)组:分离双侧颈总动脉、套线,但不阻断;脑缺血再灌注(I-R)组:阻断双侧颈 总动脉,5分钟后复灌;培高利特(I-PER)组:缺血前2天腹腔注射培高利特1mg/kg,2次/d,缺血前1小时再给 药1次;螺哌隆(I-SPI)组:腹腔注射螺哌隆1mg/kg,方法同I-PER组。Sham组和I-R组分别腹腔注射生理盐水, 容积和给药时间同I-PER组。每组分为再灌注5分钟和60分钟两个亚组。用高效液相色谱-电化学检测法测定 纹状体中DA及其代谢产物3,4-双羟苯乙酸(DOPAC)和高香草酸(HVA)的含量,计算DA代谢率([DOPAC+ HVA]/DA)。结果 I-R组再灌注5分钟和60分钟时纹状体DA含量明显低于Sham组(P均<0.01),I-PER 组、I-SPI组两时间点DA含量与I-R组相比均无显著差异(P>0.05);再灌注5分钟和60分钟时DA代谢率I-R 组明显高于Sham组(P均<0.01),I-PER组明显低于I-R组(P<0.01,P<0.05),I-SPI组明显高于I-R组(P 均<0.01)。结论 培高利特和螺哌隆可分别抑制和促进沙土鼠前脑缺血再灌注期间纹状体DA的释放和代谢。Objective To investigate the effects of pergolide and spiperone on dopamine(DA) metabolism in striatum during forebrain ischemia- reperfusion injury in gerbils. Methods Forebrain ischemia model of gerbil was produced by occlusion of bilateral carotid arteries for 5 minutes. Gerbils were randomly divided into 4 groups:Sham-operated (Sham) group, Forebrain ischemia-reperfusion (I-R) group, Pergolide (I-PER) group, Spiperone (I-SPI) group. The contents of DA,dilidroxyphenylacetic acid(DOPAC) and homovanillic acid(HVA) in striatum were measured with high performance liquid chromatography-electrochemical detection (HPLC-ECD). DA metabdic ratio([DOPAC+HVA]/DA) was calculated. Results Compared with Sham group,the DA contents in striatum decreased 26. 9%(P<0. 01) and 25. 2%(P<0. 01) respectively at 5 min and 60 min after reperfusion in I-R group. There were no statistical difference in DA contents among I-R group,I-PER group and I-SPI group;Compared with Sham group,DA metabolic ratio in I-R group increased 126. 3%(P<0. 01) and 67. 1 % (P<0. 01) respectively at 5min and 60 min after reperfusion. Compared with I-R group,DA metabolic ratio in I-PER group decreased 20. 9%(P<0. 01),18. 0%(P<0. 05) respectively,and that in I-SPI group increased 55. 8% (P<0. 01),41. 8%(P<0. 01) respectively at the two time points. Conclusion Pergolide attenuated is-chemia-reperfusion-induced decrease of DA metabolism in striatum in gerbils,while Spiperone increased it.
