小胆管上皮细胞体积量化在实验性黄疸模型中意义的探讨

摘  要:目的 探讨异硫氰酸 1-萘脂 (ANIT)所致黄疸时 ,量化小胆管上皮细胞体积的意义及与机能的关系。方法 在以ANIT所致大鼠黄疸模型上的大黄退黄疸研究中 ,通过空白组、大黄 +ANIT组和ANIT组 ,观察小胆管上皮细胞在不同条件下的体积变化 ,与显示组织细胞机能的肝肠通过时间、肝细胞摄取指数、胆汁分泌量、丙氨酸氨基转移酶活性、总胆红素和甘胆酸含量之间的关系进行了方差与逐步回归分析。结果 方差显示各项目的组间 ,除肝肠通过时间与胆汁分泌量为空白组与大黄 +ANIT组之间无差异外 ,余均有显著的统计学意义。量化的小胆管上皮细胞体积的变化与各项机能变化具有复相关 ,与肝肠通过时间和肝细胞摄取指数呈偏相关 ,有极显著的统计学意义。结论 小胆管上皮细胞的变性是ANIT所致黄疸的病理学基础 ,量化小胆管上皮细胞能以数字阐述这种组织形态变化。大黄 +ANIT组的统计学结果揭示了大黄的药理特性 ,因此试验中多角度设置项目有着重要意义。Objective To explore the relationship between volumetric changes of cholangiolar duct epithelial cells (CDEC) and their function in α-naphthylisothiocyanate(ANIT) induced jaundice. Methods The rats were assigned into three groups:one normal group,other group treated with Rhubarb and ANIT and the third group treated with only ANIT.After treatment 48hrs determined blood tests (activity of alanine aminotransferase-ALT,serum level of total bilirubin-SB and cholic acid-CA).And isotope dynamic tracing of hepatic uptake index (HI) and time needed for passing from liver to gut (HIT) were done simultaneously,together with surveying bile flow rates (BFR) and the volume of CDEC (CDECV). All the data were analysed by variance and multiple correlation of CDECV as dependent variable. Results All the blood biochemical results,the hepatic uptake index and the volume of CDECshowed significant differences between the three groups.The time needed for passing from liver to gut and the bile flow rates were of no statistical significance between normal group and the group treated with Rhubarb and ANIT, but the two groups were P<0 01 in relation to the group with only ANIT. With stepwise regression analysis of CDECV, multiple correlation coefficient was significant (P<0.05), the partial correlation coefficient of isotope dynamic tracing of HI and HIT was of highly statistical significance (P< 0 001). Conclusions The foundation of histopathological changes of jaundice caused by ANIT is degeneration of CDECV;volumetric changes of CDECV can show that histopathological change.Through the result of the group treated with Rhubarb and ANIT, pharmacology of Rhubarb is revealed.
