摘  要:目的 :用二甲基苯并蒽 (7,12 dimethylbenzanthracene ,DMBA)诱发的金黄地鼠口腔癌模型 ,研究花生四烯酸 (ArachidonicAcid ,AA)代谢相关产物在口腔癌发生过程中的作用 ,进一步探讨口腔癌的发病机制。方法 :0 .5 %DMBA涂于地鼠左侧颊囊 ,每周 3次× 6周 ,观察至 2 4周 ,用免疫组化方法检测颊黏膜组织学表现正常的黏膜、单纯增生、异常增生和癌及正常对照颊黏膜中 5 脂氧合酶 (lipoxygenase ,Lox)的表达 ;用原位杂交方法检测环氧合酶 (cyclooxygenase ,Cox) 2的表达 ;用酶联免疫方法检测口腔黏膜组织中AA相关代谢产物前列腺素(prostaglandin ,PG)E2和白三烯 (leukotriene ,LT)B4水平。 结果 :在DMBA处理的阳性对照组组织学上表现为正常的黏膜中 ,其Cox2和 5 Lox表达开始增加 ,在单纯增生、异常增生及癌组织中表达增高 ,在单纯增生及异常增生组织中Cox 2表达高于癌组织 ,且DMBA处理的口腔黏膜中PGE2和LTB4的水平显著高于未处理的口腔黏膜。结论 :AA异常代谢在口腔癌发病过程中有重要作用 ,是口腔癌发生的一个重要机制 ,抑制AA代谢可能预防口腔癌的发生。Objective:The Role of Arachidonic Acid M etabolism-associated genes a nd enzymes in DMBA-induced hamster oral carcinogenesis was studied to explore t h e carcinogenic mechanisms of oral cancer .Method: 0.5 % DMBA sol ution (in mine ral oil) was applied topically to the left cheek pouch of male Syrian golden ham sters 3 times per week for 6 weeks, and animals were observed up to the 24 th we e k. The expressions of 5- lipoxygenase(5-Lox) and cyclooxygennase-2 (Cox-2) w ere examined by immunohistochemical staining and by in situ hybridization in differ ent oral lesions including histological normal mucosa, hyperplasia, dysplasia, s quamous cell carcinoma (SCC), and normal control mucosa, respectively. In additi on, the levels of leukotriene B4 (LTB4) and Prostaglandin E2 (PGE2) were detecte d by the Enzyme immunoassay method in or without DMBA-treated oral mucosa. Result: The expressions of Cox-2 and 5-Lox were observed in the his tologically norma l areas of DMBA-treated oral tissues and increased in hyperplasia, dysplasia, a n d SCC. It appeared that the expression of Cox-2 was higher in areas with hyperp l asia, dysplasia than that in SCC. The levels of LTB4 and PGE2 were also signific antly higher in DMBA-treated oral mucosa than that without DMBA-treated oral t i ssues.Conclusion:Arachidonic Acid Metabolism plays an important role in hamster oral carcinogenesis and its inhibitors may have preventive and treatment effect s on oral cancer. [
