摘 要:目的 :研究不同亚型蛋白激酶C在肺鳞癌组织中的表达 ,以探讨它们在甲基胆蒽 (MCA)诱发的实验性肺鳞癌发生发展中可能的作用。方法 :用肺叶支气管内灌注甲基胆蒽碘油溶液诱发Wistar大鼠肺癌的方法建立大鼠肺鳞癌动物模型 ,以免疫组化方法研究PKCα、δ、ε、ξ在肺鳞癌中的表达。 结果 :用本方法诱发的肺部肿瘤均为肺鳞癌 ,诱癌率达77 8%。大鼠肺鳞癌组织中PKCα表达明显高于癌旁肺组织及正常大鼠肺组织 (分别为 2 4 . 0 8%± 2 . 93% ,5 .0 0 %± 2. 10 % ,1. 96 %±0 . 2 9% ,P <0 . 0 5 ) ,在癌旁肺组织PKCα表达也明显高于正常肺组织 (5 . 0 0 %± 2 . 10 % / 1 .96 %± 0. 2 9,P <0 . 0 5 )。与此相反 ,在肺鳞癌组织中鲜见PKCδ表达 ,癌旁肺组织中PKCδ表达明显低于正常肺组织 (分别为 4 7. 96± 6 .. 6 0 / 6 3 4 4± 6 . 4 1,P <0 . 0 5 )。在正常肺组织及肺鳞癌组织中均未见PKCε、ξ表达。结论 :诱癌过程中肺鳞癌组织PKCα表达的异常增高提示致癌剂可能通过活化PKCα介导的多个环节的信号转导而参与了肺鳞癌的发生、发展。AIM: This experiment was performed to evalu at e the significance of the expression of different types of protein kinase C (PKC ) in lung squamous cell carcinoma induced by methylcholanthrene(MAC).METHODS: Lung squamous cell carcinoma in Wistar rats was induced with MAC for 12 weeks. The stage of the cancer development was determined histop athologically. Immunohistochemistry method was used to study the expression of different types of protein kinase C α,δ,ε,ξ in lung squamous cell carcinoma. RESULTS: The level of expression of PKC α in lung squamous cel l car cinoma was significantly higher than that in peripheral lung tissues and normal lung tissues (24.08%±2.93%,5.00%±2.10%,1.96%±0.29%,respectively. P < 0.05). The expression pattern of PKC δ was reverse. There was no PKC ε and ξ found in normal lung tissues and squamous cell carcinoma.CONCLUSIONS: Our result is a strong implication that PKC α pla ys an important role in the incidence and development of the MAC-induced lung s quamous cell carcinoma through the PKC α- mediated signal transduction.
