摘  要:目的:探讨国产碱性成纤维细胞生长因子(basicfibroblastgrowthfac-tor,bFGF)对体外培养人神经干细胞增殖、分化的影响,以期寻找一种简便、经济、可靠的人神经干细胞体外培养、增殖的方法。方法:实验于2003-06/2004-03在安阳肿瘤医院临床实验室进行。机械分离8~16周胎龄水囊引产新鲜人胚胎纹状体区脑组织,用DMEM/F12无血清培养基进行培养,加不同剂量的国产bFGF刺激细胞增殖、传统方法进行传代培养,免疫组化染色对培养的细胞进行鉴定。结果:培养体系中加入终浓度28Au/mL的国产bFGF时,神经干细胞生长旺盛,形成大量悬浮状态的神经球,而且形成的神经球经多次传代仍呈悬浮球生长,绝大多数表达神经上皮干细胞蛋白(Nestin)。结论:用廉价的国产bFGF作为丝裂原体外培养人胚胎纹状体区脑组织,能获得大量神经干细胞,可以为中枢神经系统疾病神经干细胞移植治疗提供经济、可靠的细胞来源。AIM:To observe the effects of basic fibroblast growth factor(bFGF) made in China on anthropic neural stem cell(NSC) proliferation and differentiation in vitro so as to search for an economic, dependable and convenient method of anthropo neural stem cell proliferation and culture in vitro. METHODS:The experiment was conducted in Clinical Laboratory of Anyang Tumor Hospital, from June 2003 to March 2004.The fresh corpora striatum region brain tissue of 8-16 weeks fetal age anthropo embryo that were induction of labor with water bag were mechanically separated.The brain stem cells were cultivated with DMEM/F12 serum free medium,and cell proliferation was stimulated with the different dose of bFGF that made in China, and subculturing was conducted with traditional method.The cultural neural stem cells were appraised with immunohistochemistry staining. RESULTS:As to add final concentration 28 Au/mL bFGF that were made in China in culture system, the NSCs thrived,a great quantity of float neural balls were formed,and then after many times transfer of culture these neural balls were still to remain growth to offer floating balls.And the cells expressed neural epithelium stem call protein(Nestin) mostly. CONCLUSION:With cheap bFGF made in China as mitogen for in vitro culture of brain tissue in anthropo embryo corpora striatum region,we can gain generous NSCs that will provide economical and dependable cell source for NSCs transplantation therapy on the patients with central nervous system diseases.
